Abstract The tumor microenvironment harbors diverse immunosuppressive cell populations—including regulatory T cells, myeloid-derived suppressor cells, tumor-associated macrophages and other tolerogenic subsets—that drive immune evasion and therapeutic resistance. These cells are metabolically reprogrammed to sustain their suppressive function and survive under conditions of hypoxia, nutrient deprivation and oxidative stress.

Editor’s summary Lymphocytes, including regulatory T (Treg) cells, tune their metabolism to support their immunological functions. However, the molecular events controlling Treg cell metabolic reprogramming during inflammation are not fully understood. Using a mouse model of acute inflammation, Saravia et al. found that Treg cells undergo dynamic mitochondrial metabolic and lysosomal reprogramming across heterogeneous activation states.
