The RPS15-DDX21 complex drives prostate malignancy through transcriptional activation of SCD1
Abstract Development of castration resistance and distant metastasis remain two major clinical challenges in prostate cancer (PCa) treatment. By analyzing multiple public cancer datasets, we found that ribosomal protein S15 (RPS15) is overexpressed in PCa and related to its metastasis. Beyond its canonical role as a structural component of the ribosome, emerging evidence has highlighted the extraribosomal functions of RPS15 in disease progression.
WBP11 inhibits UFL1-mediated UFMylation of NONO to drive hepatocellular carcinoma progression - Oncogene
Abstract Identification of novel targets is critical for the effective management of HCC. UFMylation, a novel ubiquitin-like modification, plays an important role in various biological processes and disease progression. We found that WW domain binding protein 11 (WBP11) is enriched and predicts poor prognosis in HCC. Depletion of WBP11 inhibits the malignancy of HCC cells in vitro and in vivo.
The Ku70-SIX1-GPT2 axis regulates alpha-ketoglutarate metabolism to drive progression of prostate cancer - Oncogene
Abstract Sine oculis homeobox homolog 1 (SIX1) is a new identified cancer driver in the development of prostate cancer (PC). However, the upstream regulatory mechanisms for SIX1 reactivation in cancer remains elusive. Here, we found that Ku70 robustly interacts with SIX1 in the nucleus of PC cells. The HD domain of SIX1 and the DBD domain of Ku70 are required for formation of Ku70-SIX1 complex. 20 groups of hydrogen bonds were identified in this complex by molecular dynamics simulation.
