Abstract Vemurafenib constitutes an important therapeutic for BRAFV600 mutant melanomas, but despite high initial response rates, resistance to BRAF and MEK inhibitors quickly develops. Here, we performed an integrative analysis of metabolomic consequences and transcriptome alterations to uncover mechanisms involved in adaptive vemurafenib resistance (VemR) development and their relationship with vemurafenib tolerance thresholds.

1 Introduction Keratoconus (KC) is a progressive corneal ectatic disorder characterized by corneal thinning, irregular astigmatism, and vision deterioration (Rabinowitz, 1998). The disease is typically bilateral and usually happens in adolescence (Lucas and Burdon, 2020). It is also one of the most frequent causes of corneal transplantation (Sarezky et al., 2017).
