Hereditary alpha-tryptasemia (HαT) is a frequent tryptase gene copy number variation (TGCNV) corresponding to the presence of at least two cis-copies of the α-tryptase coding TPSAB1 gene. HαT was almost systematically associated with baseline serum tryptase levels (bST) ≥ 8 μg/L in patient-based cohorts [1].

Erdheim-Chester disease (ECD) is a rare myeloid neoplasm driven by somatic mutations in genes of the MAPK pathway [1]. The BRAFV600E mutation is found in ~60% of ECD patients and can be targeted by specific inhibitors (BRAFi), such as vemurafenib, dabrafenib, and encorafenib [1]. The introduction in 2013 of BRAFi—and more recently of MEK inhibitors (MEKi)—revolutionized the management of patients with ECD [2-4].
