CD19 CAR T cell therapy for treatment-refractory seropositive rheumatoid arthritis: a phase 1 trial
|
Abstract Chimeric antigen receptor (CAR) T cell-mediated B cell depletion has demonstrated efficacy in several autoimmune diseases. Here we report clinical and molecular data obtained during the nonrandomized phase 1 part of the phase 1/2 COMPARE trial, evaluating safety and efficacy of mivocabtagene autoleucel (miv-cel), an autologous fully human CD19 CAR T cell therapy, in rheumatoid arthritis (RA).

2023 International Rome consensus for the nomenclature of Sjögren disease - Nature Reviews Rheumatology
Abstract Nomenclature for the disease widely known as Sjögren syndrome has proven unsatisfactory. Patients have perceived ‘syndrome’ as indicative of a vague collection of symptoms, prompting the Sjögren’s Foundation to abandon the term. Furthermore, the traditional distinction between ‘primary’ and ‘secondary’ forms fails to account for the complex interplay between overlapping autoimmune diseases.

The essential roles of memory B cells in the pathogenesis of systemic lupus erythematosus - Nature Reviews Rheumatology
Abstract Emerging evidence indicates that memory B cells are dysfunctional in systemic lupus erythematosus (SLE). They are hyporesponsive to signalling through the B cell receptor (BCR) but retain responsiveness to Toll-like receptor (TLR) and type I interferon signalling, as well as to T cell-mediated activation via CD40–CD154.

Integrated Care as a Model for Interprofessional Disease Management and the Benefits for People Living with HIV/AIDS
1. Introduction In the European Union (EU), approximately 25,000 people were newly diagnosed with HIV in 2021 [1,2]. A total of 85% of newly detected cases are diagnosed at a late disease stage (defined as less than 350 CD4 cells per mm3) [3]. Compared to low-income countries, people living with HIV (PLWH) in Europe mostly have unrestricted access to antiretroviral therapy (ART).

Differences in all-cause and cause-specific mortality due to external causes and suicide between young adult refugees, non-refugee immigrants and Swedish-born young adults: The role of education and migration-related factors
Loading metrics Open Access Peer-reviewed Research Article Abstract Background International migration has increased during the past years and little is known about the mortality of young adult immigrants and refugees that came to Sweden as children.
Efficacy and Safety of the Bruton's Tyrosine Kinase Inhibitor Evobrutinib in Systemic Lupus Erythematosus: Results of a Phase II, Randomized, Double‐Blind, Placebo‐Controlled Dose‐Ranging Trial
DISCUSSION In this trial of a BTK inhibitor in patients with SLE, the primary efficacy endpoints were not met. Neither the week 52 SRI-4 responses in the mITT population nor the SRI-6 responses in the HDA subpopulation showed a clinically meaningful difference between any evobrutinib dose and placebo added on to SoC therapy.

Vaccine-Induced Thrombocytopenia with Severe Headache
Related Articles To the Editor: Vaccine-induced immune thrombotic thrombocytopenia (VITT), a serious adverse event after vaccination with ChAdOx1 nCoV-19 (AstraZeneca) or Ad26.COV2.S (Johnson & Johnson–Janssen), is caused by platelet factor 4 (PF4)–dependent, platelet-activating antibodies.1-3 High-dose immune globulins and anticoagulation are the main treatments.4,5 In this report, we present evidence that vaccine-induced thrombocytopenia (VIT) without associated cerebral venous sinus...
Health‐related and socio‐economic burden of the COVID‐19 pandemic in Vienna
Abstract Previous pandemics have rarely affected everyone equally and, so far, the COVID-19 pandemic is no exception. Emerging evidence has shown that incidence rate, hospitalisation rate, and mortality due to COVID-19 are higher among people in lower socio-economic position (SEP). In addition, first investigations indicate that not everyone is equally affected by this pandemic's collateral public health damage.
Toll-like receptor signalling in B cells during systemic lupus erythematosus
Abstract B lymphocytes have a central role in autoimmune diseases, which are often defined by specific autoantibody patterns and feature a loss of B cell tolerance. A prototypic disease associated with B cell hyperactivity is systemic lupus erythematosus (SLE). In patients with SLE, the loss of B cell tolerance to autoantigens is controlled in a cell-intrinsic manner by Toll-like receptors (TLRs), which sense nucleic acids in endosomes.
Mechanisms of action of hydroxychloroquine and chloroquine: implications for rheumatology
Abstract Despite widespread clinical use of antimalarial drugs such as hydroxychloroquine and chloroquine in the treatment of rheumatoid arthritis (RA), systemic lupus erythematosus (SLE) and other inflammatory rheumatic diseases, insights into the mechanism of action of these drugs are still emerging. Hydroxychloroquine and chloroquine are weak bases and have a characteristic ‘deep’ volume of distribution and a half-life of around 50 days.

Course and characteristics of work disability 3 years before and after lumbar spine decompression surgery- a national population-based study
Patterns of work disability were found to be very heterogeneous in patients with dorsopathy and a decompression surgery. We found five different groups of trajectories regarding SA and DP. All of them had a peak of SA/DP around the time of surgery. Still, the groups of trajectories started on different levels of SA/DP three years before surgery (ranging from zero to eight months per year).
Olumiant 4 mg efficacious for SLE
AMSTERDAM — Among patients with systemic lupus erythematosus who are receiving standard background therapy, additional treatment with daily, 4-mg doses of baricitinib was associated with significant clinical improvements compared to placebo, according to findings presented at the EULAR Annual Congress.
Efficacy of Epratuzumab, an Anti‐CD22 Monoclonal IgG Antibody, in Systemic Lupus Erythematosus Patients With Associated Sjögren's Syndrome
EMBODY 1 (ClinicalTrials.gov identifier: NCT01262365) and EMBODY 2 (ClinicalTrials.gov identifier: NCT01261793) investigated the efficacy and safety of epratuzumab, a CD22‐targeted humanized monoclonal IgG antibody, in patients with systemic lupus erythematosus (SLE). The studies showed no significant difference from placebo in primary or secondary clinical outcome measures but did demonstrate B cell–specific immunologic activity.

Unravelling the Mystery Between Structure and Sustained Clinical Outcomes
Targeted biologics have revolutionised the treatment and outlook of patients with inflammatory joint diseases. The combination of high-cost long-term therapy straining healthcare systems with impending expiry of key biologics patents has led to heightened interest in the development of biosimilars.
