The many pathways driving liver inflammation in MASH
Keywords cytokines fibrosis inflammation metabolism dysfunction-associated steatotic liver disease Introduction Metabolic dysfunction-associated steatotic liver disease (MASLD) has evolved in recent years as the most common liver disease globally.1,2,3,4 Although this disease presents in up to 80% of people as hepatic steatosis without further progression of liver disease, a substantial subgroup of patients develops metabolic dysfunction-associated steatohepatitis (MASH).5 Consequently,...

Western diets and chronic diseases - Nature Medicine
Abstract ‘Westernization’, which incorporates industrial, cultural and dietary trends, has paralleled the rise of noncommunicable diseases across the globe. Today, the Western-style diet emerges as a key stimulus for gut microbial vulnerability, chronic inflammation and chronic diseases, affecting mainly the cardiovascular system, systemic metabolism and the gut.

Heavy arch: from inflammatory bowel diseases to metabolic disorders
Conclusion The parallels and intricate link between metabolic disorders and IBD have emerged over the last decade by experimental and clinical studies alike. Current guidelines recommend treatment of obesity and metabolic disorders in patients with IBD as reported for patients without IBD. Many studies raise important questions for the future (box 3).
A guideline on the molecular ecosystem regulating ferroptosis - Nature Cell Biology
|
Abstract Ferroptosis, an intricately regulated form of cell death characterized by uncontrolled lipid peroxidation, has garnered substantial interest since this term was first coined in 2012. Recent years have witnessed remarkable progress in elucidating the detailed molecular mechanisms that govern ferroptosis induction and defence, with particular emphasis on the roles of heterogeneity and plasticity.

Diet fuelling inflammatory bowel diseases: preclinical and clinical concepts
Dietary interventions in IBD Experimental, translational and clinical evidence suggest that IBD arises from unresolved perturbation of mucosal immune responses that is determined by genetic variation and the exposome (including the diet and gut microbiota).

Cell-autonomous Hedgehog signaling controls Th17 polarization and pathogenicity - Nature Communications
Abstract Th17 cells are key drivers of autoimmune disease. However, the signaling pathways regulating Th17 polarization are poorly understood. Hedgehog signaling regulates cell fate decisions during embryogenesis and adult tissue patterning. Here we find that cell-autonomous Hedgehog signaling, independent of exogenous ligands, selectively drives the polarization of Th17 cells but not other T helper cell subsets.
Liver microbes controlling immunity: Facts and pitfalls
The microbiome in arguably sterile tissues has received considerable attention in the last few years. A recent report by Leinwand and colleagues disentangles host-microbe interactions in the mammalian liver. The authors demonstrate that a distinct liver microbiome controls hepatic immune responses in mice, challenging the perception of a sterile liver.
The intestinal microbiota fuelling metabolic inflammation
Abstract Low-grade inflammation is the hallmark of metabolic disorders such as obesity, type 2 diabetes and nonalcoholic fatty liver disease. Emerging evidence indicates that these disorders are characterized by alterations in the intestinal microbiota composition and its metabolites, which translocate from the gut across a disrupted intestinal barrier to affect various metabolic organs, such as the liver and adipose tissue, thereby contributing to metabolic inflammation.
Gut microbiota as gatekeeper of anti-tumour responses in the liver
A new report in Science by Ma and colleagues uncovers the interplay of microbiota-controlled bile acid metabolism and immune responses in the context of primary and metastatic liver tumours in mice. Their findings shed light on the gut–liver axis in hepatic malignancies.
Liver-Microbiome Axis in Health and Disease
Highlights An intestinal microbiome signature is emerging in various liver diseases independent of disease stage. Microbiota profiles may differ between various liver disease etiologies. Microbiota and microbiota-derived factors have a key role in liver cirrhosis and associated pathologies. Advanced liver disease is characterized by a circulating microbiome. Some commensals prevent liver disease in preclinical studies.
The Intestinal Microbiota in Colorectal Cancer
Experimental evidence from the past years highlights a key role for the intestinal microbiota in inflammatory and malignant gastrointestinal diseases. Diet exhibits a strong impact on microbial composition and provides risk for developing colorectal carcinoma (CRC). Large metagenomic studies in human CRC associated microbiome signatures with the colorectal adenoma-carcinoma sequence, suggesting a fundamental role of the intestinal microbiota in the evolution of gastrointestinal malignancy.
Recovery of ethanol-induced Akkermansia muciniphila depletion ameliorates alcoholic liver disease
Objective Alcoholic liver disease (ALD) is a global health problem with limited therapeutic options. Intestinal barrier integrity and the microbiota modulate susceptibility to ALD. Akkermansia muciniphila, a Gram-negative intestinal commensal, promotes barrier function partly by enhancing mucus production. The aim of this study was to investigate microbial alterations in ALD and to define the impact of A. muciniphila administration on the course of ALD.

Reversal of murine alcoholic steatohepatitis by pepducin-based functional blockade of interleukin-8 receptors
CXCR1 and CXCR2 are G protein-coupled receptors (GPCRs). 20 GPCRs consist of seven transmembrane domains joined by three intracellular loops and a C-terminal domain (referred to as i1-i4) that are important for interaction with and activation of G proteins involved in signal transduction. 21 ‘Pepducins’ are short peptides coupled to a lipid moiety that can be designed as agonists or antagonists with precise specificity in vitro and in vivo for a given GPCR.

Reversal of murine alcoholic steatohepatitis by pepducin-based functional blockade of interleukin-8 receptors
Objective Alcoholic steatohepatitis is a life-threatening condition with short-term mortality up to 40%. It features hepatic neutrophil infiltration and blood neutrophilia, and may evolve from ethanol-induced breakdown of the enteric barrier and consequent bacteraemia. Signalling through CXCR1/2 G-protein-coupled-receptors (GPCRs), the interleukin (IL)-8 receptors, is critical for the recruitment and activation of neutrophils.
