Vorasidenib improves response to subsequent chemoradiation in a genetically engineered mouse model of IDH-mutant glioma
Editor’s summary Abstract INTRODUCTION RESULTS DISCUSSION MATERIALS AND METHODS Acknowledgments Supplementary Materials REFERENCES AND NOTES Information & Authors Metrics & Citations View Options References Figures Tables Media Share Editor’s summary Gliomas with mutations in isocitrate dehydrogenase 1 or 2 (IDH1/2) are poorly understood mainly because of a lack of mutant IDH inhibitor (mIDHi)–responsive glioma models. To address this, Shi et al.

However, how αKG is supplied to these enzymes in the nucleus is unclear. Sternisha et al.

Nitrogen metabolism profiling reveals cell state-specific pyrimidine synthesis pathway choice
|
Abstract Stable isotope-tracing assays track few metabolites, yet cells use many nutrients to sustain nitrogen metabolism. Here we create a platform for tracing 30 nitrogen isotope-labelled metabolites in parallel to enable a system-level understanding of cellular nitrogen metabolism. This platform reveals that while primitive cells engage both de novo and salvage pyrimidine synthesis pathways, differentiated cells nearly exclusively salvage uridine.

Humanized neurofibroma model from induced pluripotent stem cells delineates tumor pathogenesis and developmental origins
Abstract Neurofibromatosis type 1 (NF1) is a common tumor predisposition syndrome caused by NF1 gene mutation, in which affected patients develop Schwann cell lineage peripheral nerve sheath tumors (neurofibromas). To investigate human neurofibroma pathogenesis, we differentiated a series of isogenic, patient-specific NF1-mutant human induced pluripotent stem cells (hiPSCs) into Schwannian lineage cells (SLCs).
