Th17 effector cytokines induce shared and distinct microglial and endothelial cell responses in a mouse model for post-streptococcal encephalitis
Abstract Group A Streptococcus (GAS) infections cause neuropsychiatric complications in children, but the mechanisms linking peripheral infection to brain dysfunction remain unclear. Using mouse genetics, single-cell RNA sequencing, and spatial transcriptomics, we show that GAS infections induce inflammatory transcriptional programs in microglia and brain endothelial cells (BECs), accompanied by loss of blood-brain barrier (BBB) gene expression in female mice.
Astrocyte redox imbalance underlies prelimbic neuronal hypoactivity and maladaptive affective behaviors in epilepsy

Lymphoid gene expression supports neuroprotective microglia function - Nature
Abstract Microglia, the innate immune cells of the brain, play a defining role in the progression of Alzheimer’s disease (AD)1. The microglial response to amyloid plaques in AD can range from neuroprotective to neurotoxic2. Here we show that the protective function of microglia is governed by the transcription factor PU.1, which becomes downregulated following microglial contact with plaques.
Microglia-astrocyte crosstalk regulates synapse remodeling via Wnt signaling
Keywords microglia astrocyte synapse remodeling Wnt Introduction Astrocytes and microglia are two types of non-neuronal, central nervous system (CNS)-resident cell types known to regulate synapse structure and function.1,2 Astrocytes are resident CNS glial cells that have an extensive network of fine processes that juxtapose synapses where they play critical roles in regulating neurotransmission, ion homeostasis, and synaptic plasticity.3 Astrocytes can also promote synapse formation,...

Microglia-astrocyte crosstalk regulates synapse remodeling via Wnt signaling
Abstract Astrocytes and microglia are emerging key regulators of activity-dependent synapse remodeling that engulf and remove synapses in response to changes in neural activity. Yet, the degree to which these cells communicate to coordinate this process remains an open question. Here, we use whisker removal in postnatal mice to induce activity-dependent synapse removal in the barrel cortex. We show that astrocytes do not engulf synapses in this paradigm.
A comparative analysis of microglial inducible Cre lines
Highlights • Cx3cr1CreER lines efficiently recombine but have more spontaneous recombination • Tmem119CreER, P2ry12CreER, and HexbCreER are less efficient but are microglia specific • Inter-loxP distance is a key determinant of recombination efficiency • Best practices are provided for measuring recombination in microglia Summary Cre/loxP technology has revolutionized genetic studies and allowed for spatial and temporal control of gene expression in specific cell types.
Distinct Th17 effector cytokines differentially promote microglial and blood-brain barrier inflammatory responses during post-infectious encephalitis

IL-6 boosts synaptogenesis STAT!
Maternal infection during pregnancy increases the offspring’s risk of developing neurodevelopmental disorders. While IL-6 is involved, the mechanism by which IL-6 and other cytokines affect developing neural circuits is unknown. In this issue of Immunity, Mirabella et al., 2021 Mirabella F. Desiato G.
