Peroxiredoxin 1 safeguards the nucleolar genome from oxidative damage
Abstract INTRODUCTION RESULTS DISCUSSION MATERIALS AND METHODS Acknowledgments Supplementary Materials REFERENCES Information & Authors Metrics & Citations View Options References Figures Tables Media Share This article has 0 eLetter . Abstract Peroxiredoxin 1 (PRDX1) is a highly conserved, thiol-dependent peroxidase that rapidly scavenges reactive oxygen species to modulate redox signaling.
G-quadruplex homeostasis is a determinant of PARP inhibitor toxicity in BRCA2-deficient cells
Abstract Inhibition of PARPs is a key strategy to treat tumours with defects in homologous recombination (HR), including those with mutations in the tumour suppressor gene BRCA2. PARP inhibitors generate replication stress, creating a dependence on HR to repair the resulting DNA damage. However, the DNA lesions generated upon PARP inhibition that impede replication fork progression and trigger a requirement for BRCA2 in cell survival are poorly defined.
Optical Pooled Screening for the Discovery of Regulators of the Alternative Lengthening of Telomeres Pathway
Abstract Telomere elongation is essential for the proliferation of cancer cells. Telomere length control is achieved by either the activation of the telomerase enzyme or the recombination-based Alternative Lengthening of Telomeres (ALT) pathway. ALT is active in about 10-15% of human cancers, but its molecular underpinnings remain poorly understood, preventing the discovery of potential novel therapeutic targets.

Rational optimization of a transcription factor activation domain inhibitor - Nature Structural & Molecular Biology
|
Abstract Transcription factors are among the most attractive therapeutic targets but are considered largely ‘undruggable’ in part due to the intrinsically disordered nature of their activation domains.
