Efficacy and Safety of Trifluridine/Tipiracil-Containing Combinations in Colorectal Cancer and Other Advanced Solid Tumors: A Systematic Review
Fluoropyrimidines, including 5-fluorouracil and capecitabine, alone or as part of combination regimens, have formed the mainstay in treating gastrointestinal cancers.1,2 However, resistance to fluoropyrimidines remains a considerable barrier to effective treatment.2 Trifluridine/tipiracil (FTD/TPI or TAS-102; Taiho Oncology, Inc., Princeton, NJ, USA) is an oral cytotoxic agent comprising trifluridine, a thymidine analog, and tipiracil, a thymidine phosphorylase inhibitor.3 FTD/TPI has a...
Adavosertib Enhances Antitumor Activity of Trastuzumab Deruxtecan in HER2-Expressing Cancers
This preclinical study of trastuzumab deruxtecan in combination with adavosertib demonstrates enhanced antitumor activity in HER2-expressing tumors. These findings provide supporting rationale for clinical pursuit of this combination in a select subset of patients with HER2-expressing solid tumors. Human epidermal growth factor receptor 2 (HER2) is a key oncogenic driver across several tumor types (1, 2).

ERK1/2 inhibitors act as monovalent degraders inducing ubiquitylation and proteasome-dependent turnover of ERK2, but not ERK1
The discovery that intrinsic and acquired resistance to BRAFi and/or allosteric MEKi usually arises through reactivation of ERK1/2 [11,12,15] has fuelled the development of a range of catalytic or dual-mechanism small molecule ERK inhibitors (ERKi), many of which are undergoing clinical evaluation [16,17,23]. We previously noted that ERKi treatment appeared to cause a reduction in ERK1/2 abundance [17].

The Role of cMET in Gastric Cancer—A Review of the Literature
Abstract : Simple Summary cMET is a proto-oncogene that has been extensively studied in gastric cancer. Gastric cancer (GC) is a heterogenous disease with varied histology and molecular profiling. It still implies a poor prognosis in stage IV. New targeted therapeutic options are being investigated. In this review, we analyzed all studies performed on gastric cancer with MET-inhibitors. In first-line therapy, the addition of MET-inhibition to chemotherapy did not show any benefit in allcomers.

How to Tackle Mental Fatigue: A Systematic Review of Potential Countermeasures and Their Underlying Mechanisms - Sports Medicine
Abstract Introduction Mental fatigue (MF) is a psychobiological state that impairs cognitive as well as physical performance in different settings. Recently, numerous studies have sought ways to counteract these negative effects of MF. An overview of the explored countermeasures for MF is, however, lacking. Objectives The objective of this review is to provide an overview of the different MF countermeasures currently explored in literature.
Adult Female Sleep During Hypoxic Bed Rest
Introduction Maintaining a hypobaric hypoxic environment within a spacecraft or planetary habitat is being considered as a potential solution to minimise decompression procedures in preparation for extravehicular activities (EVAs), and extrahabitat activities on the Moon and Mars (Norcross et al., 2015). The boundary conditions for such future ambients have not yet been resolved.
The KEYNOTE-811 trial of dual PD-1 and HER2 blockade in HER2-positive gastric cancer - Nature
Abstract Human epidermal growth factor receptor 2 (HER2, also known as ERBB2) amplification or overexpression occurs in approximately 20% of advanced gastric or gastro-oesophageal junction adenocarcinomas1,2,3. More than a decade ago, combination therapy with the anti-HER2 antibody trastuzumab and chemotherapy became the standard first-line treatment for patients with these types of tumours4.
GATA6 Expression Distinguishes Classical and Basal-like Subtypes in Advanced Pancreatic Cancer
Combination chemotherapy is used in the treatment of most patients with advanced PDAC, yet the field is lacking robust biomarkers of outcome to guide regimen selection. Here, we show that patients with tumors of a modified “basal-like” phenotype, or those with low GATA6 RNA expression, have inferior outcomes compared with those with the “classical” phenotype. The latter are accurately identified by high GATA6 expression and positive GATA6 staining by ISH.
