Tackling aging and chronic disease: How space exploration and the ‘chameleon effect’ of pharmaceuticals can lead the way
1 NECROSIS VERSUS PROGRAMMED CELL DEATH Cells are the core building blocks of life. Their death can be either a carefully orchestrated process to maintain survival (programmed cell death) or the defining driver of biological degeneration (necrosis). Necrosis is distinct from programmed forms of cell death, such as apoptosis, autophagy, necroptosis and ferroptosis, which are genetically regulated and play adaptive roles in maintaining tissue homeostasis and preventing diseases such as cancer.
Computer Science > Computation and Language
Large language models (LLMs) have recently shown strong performance on Theory of Mind (ToM) tests, prompting debate about the nature and validity of the underlying capabilities. At the same time, reasoning-oriented LLMs trained via reinforcement learning with verifiable rewards have demonstrated notable improvements across a range of benchmarks.

Usability and feasibility assessment of the Huntington Support App (HD-eHelp study): a mixed-methods multi-study report - BMC Digital Health
Abstract eHealth holds great promise for managing rare and complex neurodegenerative diseases, such as Huntington’s disease (HD). To address this potential, the Huntington Support App (HSA) was developed to provide reliable information on HD, news updates, and contact possibilities tailored primarily to the needs of HD gene expansion carriers (HDGECs) and their partners.

Activated αβ T- and reduced mucosa-associated invariant T cells in LGI1- and CASPR2-encephalitis
Anti-Leucine-rich glioma inactivated-1 (LGI1) and anti-contactin-associated-protein-2 (CASPR2) autoimmune encephalitis (AIE) are common and characterized by pathogenic antibodies targeting neuronal autoantigens. However, the drivers of the antibody-secreting cells (ASC) and involvement of T cells remain unresolved.
Mendelian randomization identifies proteins involved in neurodegenerative diseases
Neurological diseases are the leading cause of disability and the second leading cause of death worldwide.1 Neurodegenerative diseases constitute a distinct group of neurological diseases, which are characterized by progressive neuronal loss and formation of distinct pathological changes in the brain.2 During the last three decades, there has been a substantial increase in the number of people living with neurodegenerative diseases such as Alzheimer’s disease (AD), Parkinson’s disease (PD),...

Association of oxidative stress and inflammatory metabolites with Alzheimer’s disease cerebrospinal fluid biomarkers in mild cognitive impairment - Alzheimer's Research & Therapy
Study populations Study participants included in the analyses came from two cohorts of the Alzheimer's Disease Apolipoprotein Pathology for Treatment Elucidation and Development (ADAPTED) consortium, including Barcelona-based memory clinic Ace Alzheimer Center Barcelona (147 CSF-plasma paired samples) and Heidelberg/Mannheim memory clinic (58 CSF samples). Demographic information of the full data set is provided in Supplementary Table 1.
Association of oxidative stress and inflammatory metabolites with Alzheimer’s disease cerebrospinal fluid biomarkers in mild cognitive impairment - Alzheimer's Research & Therapy
Abstract Isoprostanes and prostaglandins are biomarkers for oxidative stress and inflammation. Their role in Alzheimer's disease (AD) pathophysiology is yet unknown. In the current study, we aim to identify the association of isoprostanes and prostaglandins with the Amyloid, Tau, Neurodegeneration (ATN) biomarkers (Aβ-42, p-tau, and t-tau) of AD pathophysiology in mild cognitive impairment (MCI) subjects.
Immunogenicity of 2 therapeutic mosaic HIV-1 vaccine strategies in individuals with HIV-1 on antiretroviral therapy - npj Vaccines
Abstract Mosaic HIV-1 vaccines have been shown to elicit robust humoral and cellular immune responses in people living with HIV-1 (PLWH), that had started antiretroviral therapy (ART) during acute infection. We evaluated the safety and immunogenicity of 2 mosaic vaccine regimens in virologically suppressed individuals that had initiated ART during the chronic phase of infection, exemplifying the majority of PLWH.
Plasma trimethylamine N-oxide (TMAO): associations with cognition, neuroimaging, and dementia - Alzheimer's Research & Therapy
This study was embedded in the Rotterdam Study (RS), with a partial validation for neuroimaging associations in the Leiden Longevity Study (LLS). A brief description of both cohorts can be found in Methods S1 [14, 15]. Both studies comply with Declaration of Helsinki and are approved by the Medical Ethical Committee of the respective institutional review boards (registration number MEC 02.1015 and P01.113). All participants provided written informed consent.
Assessing the quality of chest compressions with a DIY low-cost manikin (LoCoMan) versus a standard manikin: a quasi-experimental study in primary education | European Journal of Pediatrics
Abstract Extending the access to cardiopulmonary resuscitation (CPR) training to a wider public is an important step in increasing survivability of out-of-hospital cardiac arrest. However, often price and maintenance of CPR manikins are barriers that prevent training at schools.

Germline mutation rate is elevated in young and old parents in Caenorhabditis remanei
Germline mutations are heritable DNA alterations in the germ cells that can be transmitted to future generations, and therefore are the ultimate source of genetic diversity and the fuel for evolution. The rate at which germline mutations arise is central to fields ranging from evolutionary biology to clinical research.
A type 1 diabetes polygenic score is not associated with prevalent type 2 diabetes in large population studies
Both type 1 (T1D) and type 2 (T2D) diabetes have significant genetic contributions to risk and understanding their overlap can offer clinical insight. We examined whether a T1D polygenic score (PS) was associated with a diagnosis of T2D in the Cohorts for Heart and Aging Research in Genomic Epidemiology (CHARGE) consortium. We constructed a T1D PS using 79 known single nucleotide polymorphisms associated with T1D risk.
Single-cell profiling reveals age-associated immunity in atherosclerosis
Aging is a dominant driver of atherosclerosis and induces a series of immunological alterations, called immunosenescence. Given the demographic shift towards elderly, elucidating the unknown impact of aging on the immunological landscape in atherosclerosis is highly relevant. While the young Western diet-fed Ldlr-deficient (Ldlr-/-) mouse is a widely used model to study atherosclerosis, it does not reflect the gradual plaque progression in the context of an aging immune system as occurs in humans.

Using the embryo-uterus statistical model to predict pregnancy chances by using cleavage stage morphokinetics and female age: two centre-specific prediction models and mutual validation | Reproductive Biology and Endocrinology | Full Text
Here, we aimed to develop a TLM prediction model that is able to predict pregnancy chances after SET and DET. This work resulted in two centre-specific prediction models that predict the chance for achieving a pregnancy based on a limited number of morphokinetic parameters and female age. Our used methods are new in the field and add another perspective to handling the data generated by time-lapse incubators and how this information could be useful for decision making.

Variant of TREM2 Associated with the Risk of Alzheimer's Disease
50 References 916 Citing Articles Letters Abstract Background Sequence variants, including the ε4 allele of apolipoprotein E, have been associated with the risk of the common late-onset form of Alzheimer's disease. Few rare variants affecting the risk of late-onset Alzheimer's disease have been found. Methods We obtained the genome sequences of 2261 Icelanders and identified sequence variants that were likely to affect protein function.
