Generation of Pum1 and/or Pum2 knockout mESCs Paired guide RNAs (gRNAs) targeting specific sequences within exon 12 of Pum1 and exon 6 of Pum2, with a spacing of 5–30 bp (Figure S1A), were designed using the Benchling-sgRNA online tool (https://www.benchling.com/crispr/). Each sgRNA pair was cloned into a pGL3-U6-sgRNA-PGK-puromycin vector (Addgene plasmid Cat# 51133).
