Probabilistic Fine-mapping of Putative Causal Genes
Abstract Integrative genetic analysis of molecular and complex trait data, including colocalization analysis and transcriptome-wide association studies (TWAS), has shown promise in linking GWAS findings to putative causal genes (PCGs) underlying complex diseases. However, existing methods have notable limitations: TWAS tend to produce an excess of false-positive PCGs, while colocalization analysis often lacks sufficient statistical power, resulting in many false negatives.
