Overexpression of KRAS-Q25A mutant with enhanced GTPase activity inhibits the progression of KRAS-G12D colorectal cancer
Abstract Oncogenic KRAS mutations occur in approximately 40% of colorectal cancer (CRC) cases and correlate with poor prognosis. While restoring GTP hydrolysis in KRAS-mutant tumors represents a promising therapeutic strategy, its mechanistic basis remains unclear. KRASQ25A mutant exhibits enhanced intrinsic GTPase activity, but its cellular impact is unknown. Here, we investigated the tumor-suppressive effects of KRASQ25A in KRASG12D-mutant CRC.
Reproductive behaviors, genetic susceptibility and accelerated aging risk
Abstract We examined the associations of reproductive behaviors and genetic susceptibility with aging indicators among female participants in the UK Biobank. Reproductive behaviors included the number of children ever born (NEB), age at first birth (AFB), age at last birth (ALB), and span of years of births (SYB). Aging indicators included frailty, PhenoAge acceleration, KDM-BA acceleration, and brain age difference (BrainAGE-diff).
CD8 + T cell loss induces autoinflammation in inborn errors of cell death

Abstract Systemic autoinflammatory diseases (SAID) with inborn errors of cell death (IECD) are caused by overactivation of programmed cell death (PCD). However, the pathogenesis by which PCD leads to autoinflammation remains unclear. Here, we identified IECD patients carrying compound heterozygous RIPK1 variants K377E/R390G with autoinflammatory manifestations. Mechanistically, K377E and R390G mutations suppress NF-κB signaling and activate RIPK1 to promote cell death.
