Single-cell mRNA sequencing (scRNA-seq), a powerful tool for defining the cellular diversity in multicellular organisms by parsing differential gene expression, has been applied in a variety of studies. (1−4) However, the variation between single cells cannot be fully understood by mRNA profiling alone, since post-transcriptional regulation plays a pivotal role in shaping single-cell heterogeneity.

Mammalian cell culture Human K562 cells (ATCC, CCL-243) and human colorectal cancer HCT116 cells (ATCC, CCL-247) were cultured in high glucose Dulbecco’s modified Eagle’s medium (DMEM) supplemented with 10% fetal bovine serum (Gibco, catalog no. 11965) and 1× penicillin-streptomycin (Gibco, catalog no. 15140122; 100 U/mL of penicillin, 100 µg/mL of streptomycin) and maintained at 37 °C with 5% CO2.
