Author Correction: Engineered CRISPR-OsCas12f1 and RhCas12f1 with robust activities and expanded target range for genome editing
Correction to: Nature Communications https://doi.org/10.1038/s41467-023-37829-7, published online 11 April 2023 In the version of this article initially published, replicate values were inadvertently duplicated in Figs. 4c and 5c and the associated Source Data. Consequently, the summary values in Fig. 4d were affected by the error in Fig. 4c. In addition, the values for NLRC4 OT5, intergenic OT4 and CLIC4 OT4 in Fig. 5c were presented as percentages without being multiplied by 100.
Repurposing Type I-A CRISPR-Cas3 for a robust diagnosis of human papillomavirus (HPV) - Communications Biology
Abstract R-loop-triggered collateral single-stranded DNA (ssDNA) nuclease activity within Class 1 Type I CRISPR-Cas systems holds immense potential for nucleic acid detection. However, the hyperactive ssDNase activity of Cas3 introduces unwanted noise and false-positive results. In this study, we identified a novel Type I-A Cas3 variant derived from Thermococcus siculi, which remains in an auto-inhibited state until it is triggered by Cascade complex and R-loop formation.
Development of deaminase-free T-to-S base editor and C-to-G base editor by engineered human uracil DNA glycosylase - Nature Communications
Abstract DNA base editors enable direct editing of adenine (A), cytosine (C), or guanine (G), but there is no base editor for direct thymine (T) editing currently. Here we develop two deaminase-free glycosylase-based base editors for direct T editing (gTBE) and C editing (gCBE) by fusing Cas9 nickase (nCas9) with engineered human uracil DNA glycosylase (UNG) variants.
