Gα o/z -biased coupling profile of the dopamine D3 receptor
Abstract Dopamine receptors are G protein coupled receptors (GPCRs) that serve as key targets for FDA-approved drugs to treat various neuropsychiatric disorders. Notably, ~11% of all marketed GPCR-targeting drugs act on dopamine receptors. Five GPCRs mediate the effects of endogenous dopamine and compounds used to treat Parkinson’s disease, schizophrenia, and other conditions.
Pathogenic variants in KCTD1 disrupt cAMP signaling and cellular communication associated with developmental pathways
Keywords Potassium channel tetramerization domain 1 (KCTD1) G protein‐coupled receptor (GPCR) cell signaling signal transduction cyclic AMP (cAMP) adenylate cyclase type 5 (AC5) KCTD Wnt Notch Hh Introduction Potassium Channel Tetramerization Domain-containing (KCTD) proteins consist of 25 members hallmarked by a common bric-à-brac, tramtrack, broad complex (BTB) domain, which endows their ability to form interactions with numerous proteins (1, 2).
The G protein modifier KCTD5 tunes the decoding of neuromodulatory signals necessary for motor function in striatal neurons
Methods Experimental protocols were approved by Augusta University’s Institutional Animal Care and Use Committee (Animal Use Protocol 2020-1036) and Institutional Biosafety Committee (Biosafety Protocol #2037). Mice were housed under standard conditions on a 12-hour light/dark cycle with unlimited access to both food and water.
