CUT&Tag The CUT&Tag sequencing was performed with the CUT&Tag-IT Assay Kit (Active motif, 53160) by following the manufacturer's protocol. Briefly, 105 OVCAR5 ALDH+ CSCs and OVCAR5 ALDH− non-CSCs were FACS-sorted and incubated with Concanavalin A beads for binding. Cells were then incubated with a primary antibody targeting H3K27me3 (Active Motif, 39155), H3K4me3 (Abcam, ab8580), and H3K27ac (Active Motif, #39034) overnight at 4 °C.

1 Introduction Emergence of platinum (Pt) resistance (Pt-R) predicts invariably fatal outcomes in high grade serous ovarian cancer (HGSOC) with an expected median survival of 12–18 months.[1, 2] Pt-R HGSOC remains an area of unmet need, where progress has remained slow and clinical outcomes are lagging.[3-6] Understanding key vulnerabilities of Pt-R OC could unlock new possibilities to target HGSOC and improve survival.

Abstract Following a period of slow progress, the completion of genome sequencing and the paradigm shift relative to the cell of origin for high grade serous ovarian cancer (HGSOC) led to a new perspective on the biology and therapeutic solutions for this deadly cancer. Experimental models were revisited to address old questions, and improved tools were generated.
