1 Introduction Allogeneic hematopoietic stem cell transplantation (HSCT) is a standard of care treatment for a range of hematological malignancies. Chronic GVHD (cGVHD) and the complications of its treatment are among the leading causes of morbidity and mortality following HSCT [1-3]. Advances have been made in cGVHD prevention, most significantly in the areas of conditioning and prophylaxis regimens, but incidence remains high at 30%–50% [4].

Abstract CD19-directed chimeric antigen receptor (CAR)-T cells are breakthrough therapies for aggressive B-cell lymphomas, but less than half of patients achieve durable responses. We previously showed through whole-genome sequencing of tumors from CAR-T-treated patients that deletions of RHOA (3p21.31) are enriched in cases progressing after treatment.
