Deficiency of non-muscle myosin IIA promotes neuroinflammation by impairing autophagic degradation of VCAM-1 in brain endothelium
Abstract Endothelial cell dysfunction is a major driver of neuroinflammation, a critical contributor to neurological disorders. Non-muscle myosin IIA (NMIIA), a key cytoskeletal protein, has been implicated in cerebrovascular pathology, but its role in endothelial-mediated neuroinflammation remains unclear. This study reveals that NMIIA expression is downregulated in brain microvascular endothelial cells during aging.
Venetoclax, azacitidine, and G-CSF targeting molecular residual disease in CBFB::MYH11 AML resistant to high-dose cytarabine
|
Dear Editor, Acute myeloid leukemia (AML) with the CBFB::MYH11 rearrangement is generally considered a favorable-risk subtype highly responsive to intensive cytarabine-based chemotherapy [1]. However, up to 30% of patients ultimately relapse, driven largely by persistent measurable residual disease (MRD) [1].

Caspases and programmed cell death in sepsis: mechanisms, pathophysiology, and therapeutic targets
Abstract Sepsis represents a critical medical condition characterized by organ impairment resulting from an uncontrolled systemic reaction to pathogenic infection, posing substantial difficulties in intensive care management. As a key pathological mechanism, programmed cell death (PCD) critically contributes to sepsis development, and caspases act as the central molecular hub governing multiple PCD modalities, including apoptosis, necroptosis, pyroptosis, and PANoptosis.

Molecular insights into ligand recognition and signaling of OXGR1 - Nature Communications
Abstract GPR99/OXGR1 is a G protein-coupled receptor (GPCR) with two endogenous agonists, the tricarboxylic acid cycle derivative 2-oxoglutarate (α-ketoglutarate) and the inflammatory mediator cysteinyl leukotriene E4 (LTE4), hence also termed CysLT3 receptor. How GPR99/OXGR1 recognizes two distinct ligands is a biologically important question. Here we present cryo-EM structures of GPR99/OXGR1-Gq complexed with oxoglutarate and LTE4, respectively.
A Metal-free Molecular Pyroelectric Material with Strong Pyro-Photovoltaic Coupling for Enhancing Self-Powered X-ray Response Metal-free X-ray detectors have shown enormous application potential in the fields of public safety and medical diagnostics, owing to their environmentally sustainable and low-cost characteristics. However, the X-ray absorption and response capabilities of metal-free materials are constrained by their intrinsic low atomic number and low density.
Correlation and prognosis analysis of human papillomavirus infection and P16 expression in oral and oropharyngeal squamous cell carcinomas - Scientific Reports
Abstract The incidence of HPV-related oral squamous cell carcinomas (OSCC)and oropharyngeal squamous cell carcinomas (OPSCC) has increased significantly in recent years, but the role of HPV and P16 in OSCC and OPSCC remains controversial. Here, we evaluate the prevalence and prognostic significance of HPV-DNA, HPV E6E7 and P16 protein expression in OSCC and OPSCC patients. Additionally, we explore the correlation between P16 protein expression and HPV infection in these cases.
Structural basis for full-length chemerin recognition and signaling through chemerin receptor 1 - Communications Biology
Abstract Chemerin, a chemotactic adipokine, plays essential roles in adipogenesis and inflammation. Serum chemerin concentration is closely associated with obesity and metabolism disorders. The mature form of chemerin (residues 21-157) acts primarily through chemerin receptor 1 (CMKLR1) for transmembrane signaling. As a result, CMKLR1 serves as a promising target for therapeutic intervention of immunometabolic diseases such as diabetes and multiple sclerosis.

Effective cervical intraepithelial neoplasia classification through polarization-based machine learning
Identification of cervical intraepithelial neoplasia (CIN) tissues holds significant clinical importance in reducing the incidence and mortality of cervical cancer. The ultraviolet Mueller matrix imaging polarimeter (UV-MMIP) can significantly enhance morphological specific contrast. For the first time, to our best knowledge, we used polarimetric metrics measured by UV-MMIP combined with machine-learning techniques to achieve high-accuracy CIN classification.

Structural insights into agonist binding and activation of succinate receptor 1
Abstract Succinate is an intermediate of the citric acid cycle and serves important functions in energy homeostasis and metabolic regulation. Extracellular accumulation of succinate acts as a stress-induced signal through its G protein-coupled receptor, SUCNR1. Research on succinate signaling is hampered by the lack of high-resolution structures of the agonist-bound receptor.

Structural basis for constitutive activation and CXCL1 recognition of human herpesvirus 8-encoded G protein-coupled receptor KSHV-GPCR
Abstract Kaposi's sarcoma-associated herpesvirus (KSHV) encodes a viral G protein-coupled receptor, KSHV-GPCR, that contributes to KSHV immune evasion and pathogenesis of Kaposi's Sarcoma. KSHV-GPCR shares a high similarity with CXC chemokine receptors CXCR2 and can be activated by selected chemokine ligands. KSHV-GPCR is also unique for its constitutive activity by coupling to various G proteins.
Cryo-EM structure of monomeric CXCL12-bound CXCR4 in active state
Abstract CXC chemokine receptor 4 (CXCR4) and its chemokine ligand CXCL12 are crucial to embryonic development, bone marrow retention of hematopoietic progenitor cells, cancer metastasis, angiogenesis and HIV-1 infection. Yet the structural basis for CXCR4 recognition of full-length CXCL12 remains unknown despite available structures of CXCR4 in complex with small molecule antagonists and a viral chemokine.

Cryo-EM structures of the human G-protein coupled receptor 1 (GPR1) - Gi protein complexes bound to the full-length chemerin and to its C-terminal nonapeptide | bioRxiv
Abstract Chemerin is an adipokine with chemotactic activity to a subset of leukocytes. Chemerin mainly acts through its C-terminal nonapeptide (YFPGQFAFS, C9) that bind to three G protein-coupled receptors including chemokine-like receptor 1 (CMKLR1), G-protein coupled receptor 1 (GPR1) and C-C chemokine receptor-like 2 (CCRL2). We examined C9 signaling through GPR1 and found this receptor capable of Gi signaling but very weak β-arrestin signaling.
Structural basis for EROS binding to human phagocyte NADPH oxidase NOX2
Abstract EROS (essential for reactive oxygen species) is a recently identified molecular chaperone of NOX2 (gp91phox), the catalytic subunit of phagocyte NADPH oxidase. Deficiency in NOX2 expression or function due to genetic mutations leads to chronic granulomatous disease (CGD) with recurrent bacterial and fungal infections. To delineate how EROS interacts with NOX2, we solved the cryo-EM structure of the EROS-NOX2-p22phox heterotrimeric complex.
