We noticed that the Subdued signal was also found within the midgut visceral muscles. Analysis of confocal stack images of midguts co-stained with phalloidin unmasked its expression specifically to the circular muscles, while, as mentioned above, virtually completely lacking from longitudinal ones (Figures 4A, 4B and 4C; Figure S5C, S5D and S5E; compare RGECO signal in Figure 4D), further confirming our initial observations based on mef2>RGECO movies.
