A conserved GTPase complex gates exocytosis for defence and parasitism across alveolates
Abstract Regulated protein secretion is a fundamental process by which eukaryotic cells respond to and interact with their dynamic environments, yet its mechanism outside metazoans remains largely unknown. Here we uncover an atypical small GTPase complex (Nd-GTPase–Nd9–NdP1) conserved across the Alveolata—a major eukaryotic clade encompassing ciliates and apicomplexan parasites—that gates organelle exocytosis for diverse functions ranging from cellular defence to host invasion.

Defining How Oncogenic and Developmental Mutations of PIK3R1 Alter the Regulation of Class IA Phosphoinositide 3-Kinases
Highlights • Oncogenic Q572∗ variant of p85α leads to hyper-activation of p110α compared with p110δ • HDX-MS revealed that Q572∗ leads to disruption of all inhibitory interfaces in p110α • An SHORT syndrome mutation in p85α leads to decreased sensitivity to RTKs for p110α/δ Summary The class I phosphoinositide 3-kinases (PI3Ks) are key signaling enzymes composed of a heterodimer of a p110 catalytic subunit and a p85 regulatory subunit, with PI3K mutations being causative of multiple human...
How do you tell a colleague that they have a lousy startup idea?
It rarely works to tell someone their idea is bad. Besides, how do you know? But you might know if the market for it is bad. First, I suggest you ask them whether they are interested in hearing input. Then, ask whether they want polite, supportive input, or sincere, critical, evaluative, potentially helpful input that may sound harsh. If they don't welcome the second option, disengage. You will be wasting your time.

Dissecting the molecular assembly of the Toxoplasma gondii MyoA motility complex
Apicomplexan parasites such as Toxoplasma gondii rely on a unique form of locomotion known as gliding motility. Generating the mechanical forces to support motility are divergent class XIV myosins (MyoA) coordinated by accessory proteins known as light chains. Although the importance of the MyoA–light chain complex is well-established, the detailed mechanisms governing its assembly and regulation are relatively unknown.
