FBL facilitates interstrand DNA crosslink repair by 2’-O-methylating FANCD2 mRNA
Abstract DNA interstrand cross-links (ICLs) are highly cytotoxic DNA lesions implicated in cancer and degenerative diseases. The Fanconi anemia (FA) pathway is the primary repair mechanism for ICLs, with FANCD2 monoubiquitination being crucial for the activation of this pathway. However, the role of epitranscriptional regulation in ICL repairs remains poorly understood. In this study, we demonstrate that the 2’-O-methyltransferase fibrillarin (FBL) is downregulated in FA patients.
Mutant TP53 hijacks RNA-splicing factor RBM28 to suppress double-stranded RNA triggered antitumor immunity
Abstract TP53 mutation may not only compromise its multifaceted tumor-suppressive functions but also confer oncogenic properties. Here, we demonstrate that DNA-binding domain mutations of TP53 unexpectedly confer a transcriptional regulatory function, directly driving RNA-splicing factor RBM28 overexpression.

Krüppel-like factor 4 downregulation during esophageal cancer formation facilitates immune evasion and impairs immunotherapy
Abstract Impaired antigen processing and presentation are critical for cancer immune evasion, but the underlying mechanism is not fully known. Here, we show that downregulation of Krüppel-like factor 4 (KLF4) expression during the development of cancer is essential in this immuno-decisive process. We find that suppressed KLF4 expression causes diminished MHC class I production in esophageal cancer, forming immunosuppressive niches that limit CD8⁺ T cell infiltration.

CKS1B is a tumor-intrinsic factor driving CD8 + T cell exhaustion via maintaining persistent tumor-antigen stimulation
Abstract T cell exhaustion is a major barrier to effective antitumor immunity, yet the tumor-intrinsic mechanisms remain poorly defined. Through single-cell and spatial proteomics analyses of esophageal squamous cell carcinoma (ESCC), we uncover two infection-like CD8+ T cell trajectories, acute-like and chronic-like responses, whose fates are dictated by the tumor cell subtypes they encounter. This concept links tumor heterogeneity to the shaping of local immune niches.
