Structural and functional characterization of a conserved cryptic epitope on SARS-CoV-2 spike S2 subunit

HEK293T cells were harvested 48 h post-transfection and washed once with PBS before gentle resuspension in PBS containing 1% BSA and 2 mM EDTA at pH 7.4 (PBS-BSA). Cell density was measured using a LUNA-II Automated Cell Counter (Logos Biosystems, Cat. No. L40002), and cells were centrifuged at 200 × g for 1 min. After decanting the supernatant, cells were resuspended at 3 × 106 cells/mL in chilled PBS-BSA.

Abstract Ever-evolving SARS-CoV-2 variants of concern (VOCs) have diminished the effectiveness of therapeutic antibodies and vaccines. Developing a coronavirus vaccine that offers a greater breadth of protection against current and future VOCs would eliminate the need to reformulate COVID-19 vaccines. Here, we rationally engineer the sequence-conserved S2 subunit of the SARS-CoV-2 spike protein and characterize the resulting S2-only antigens.

Abstract The ongoing evolution of SARS-CoV-2 into more easily transmissible and infectious variants has provided unprecedented insight into mutations enabling immune escape. Understanding how these mutations affect the dynamics of antibody-antigen interactions is crucial to the development of broadly protective antibodies and vaccines. Here we report the characterization of a potent neutralizing antibody (N3-1) identified from a COVID-19 patient during the first disease wave.
