An inappropriate decline in ribosome levels drives a diverse set of neurodevelopmental disorders
Abstract Many neurodevelopmental defects are linked to perturbations in genes involved in housekeeping functions, such as those encoding ribosome biogenesis factors. However, how reductions in ribosome biogenesis can result in tissue and developmental specific defects remains a mystery. Here we describe new allelic variants in the ribosome biogenesis factor AIRIM primarily associated with neurodevelopmental disorders.

Labeling of heterochronic ribosomes reveals C1ORF109 and SPATA5 control a late step in human ribosome assembly
Highlights • Ribo-SNAP enables live tracking of old and new ribosomes at single-cell resolution • C1ORF109, CINP, SPATA5, and SPATA5L1 are essential for ribosome maturation • Microcephaly associated SPATA5 mutations affect ribosome biogenesis Summary Although features of ribosome assembly are shared between species, our understanding of the diversity, complexity, dynamics, and regulation of ribosome production in multicellular organisms remains incomplete.
