Oral 4′-fluorouridine rescues nonhuman primates from advanced Lassa fever - Nature

Abstract There are no approved treatments for Lassa fever, which is estimated to cause 100,000 to 300,000 infections and 5,000 deaths annually in West Africa1,2. Recently, it was shown that 4′-fluorouridine (also known as EIDD-2749), an orally available ribonucleoside analogue, protected guinea pigs from lethal challenge with the lineage IV prototype Josiah strain of Lassa virus when treatment was delayed beyond the onset of clinical signs of disease3.

Results Historically, most CMs used for LASV studies have originated from Asian countries other than Mauritius as noted earlier, which are currently in short supply. To compare the pathogenic potential of LASV when administered i.n. as atomized particles, we challenged a total of 10 Mauritius-origin cynomolgus macaques (MCMs) and 10 AGMs with a 1,000 plaque-forming unit (PFU) of a contemporary LASV lineage II isolate (0043/LV/14).

Abstract In 2018, a clinical trial of four investigational therapies for Ebola virus disease (EVD), known as the PALM trial, was conducted in the Democratic Republic of Congo. All patients received either the antiviral remdesivir (RDV) or a monoclonal antibody product: ZMapp, mAb114 (Ebanga), or REGN-EB3 (Inmazeb). The study concluded that both mAb114 and REGN-EB3 were superior to ZMapp and RDV in reducing mortality from EVD.
