Abstract Regulatory factor X 7 (RFX7) nonsense mutations have been found in different human B cell malignancies. We therefore set out to study the role of RFX7 in B cell activation and lymphomagenesis. Here we show that RFX7 truncations cause loss-of-function and dominant-negative effects. Moreover, low RFX7 mRNA levels correlate with worse diffuse large B cell lymphoma prognosis.

Injection of chimeric chemokines reveals expression of ACKRs in the kidney We combined the use of the ACKR reporter mice and the chimeric chemokines that we produced to study expression of ACKR3, ACKR4, and GRP182 in the kidney. By using double reporter ACKR3-GFP–GPR182-mCherry, we were able to identify expression of GPR182 in primarily in glomeruli and in the vascular pole of the Bowman’s capsule (Fig 8A). Previous reports [54] identified GPR182 in podocytes.

Highlights • ACKR3 marks two equal-sized marginal zone B cell (MZB) populations • ACKR3-negative MZBs can differentiate into ACKR3-positive cells, but not vice versa • Transferred ACKR3ko follicular B cells fail to populate an empty marginal zone • ACKR3-deficient MZBs are impaired in mounting an early immune response Summary The marginal zone (MZ) contributes to the highly organized spleen microarchitecture.
