A case of Bundibugyo virus disease treated with monoclonal antibodies and remdesivir

Abstract Clinical data on Bundibugyo virus (BDBV) infection are limited, and no approved virus-specific treatment exists. We describe the case of a previously healthy 39-year-old healthcare worker who acquired BDBV infection while working in Ituri Province, Democratic Republic of the Congo (DRC), and was medically evacuated to Germany.

To develop an HIV-1 cure, insight into immunological mechanisms capable of preventing HIV-1 viral rebound is urgently needed. Here, we describe three exceptional post-intervention controllers (PICs) who maintained ART-free virological control for >6.5 years (ongoing), >7.5 years (ongoing) and 2.5 years following administration of broadly neutralizing antibodies.

Here we characterize a case of long-term HIV-1 remission of a 53-year-old male who was carefully monitored for more than 9 years after allogeneic CCR5Δ32/Δ32 HSCT performed for acute myeloid leukemia.

Antibody Teamwork against Ebola Virus Disease
Antibodies are key components to prevent and treat Ebola virus disease (EVD). In this issue of Immunity, Gilchuk et al. decipher the underlying mechanism of two antibodies that cooperatively target Ebolaviruses. The study provides insight into an antibody combination that potentiates antiviral activity and is able to prevent EVD in nonhuman primates.

Abstract Combination antiretroviral therapy (ART) is highly effective in controlling human immunodeficiency virus (HIV)-1 but requires lifelong medication due to the existence of a latent viral reservoir1,2. Potent broadly neutralizing antibodies (bNAbs) represent a potential alternative or adjuvant to ART. In addition to suppressing viremia, bNAbs may have T cell immunomodulatory effects as seen for other forms of immunotherapy3.

Abstract Recombinant vesicular stomatitis virus–Zaire Ebola virus (rVSV-ZEBOV) is the most advanced Ebola virus vaccine candidate and is currently being used to combat the outbreak of Ebola virus disease (EVD) in the Democratic Republic of the Congo (DRC). Here we examine the humoral immune response in a subset of human volunteers enrolled in a phase 1 rVSV-ZEBOV vaccination trial by performing comprehensive single B cell and electron microscopy structure analyses.
Broadly neutralizing anti-HIV-1 monoclonal antibodies in the clinic
Abstract Combination anti-retroviral therapy (ART) has revolutionized the treatment and prevention of HIV-1 infection. Taken daily, ART prevents and suppresses the infection. However, ART interruption almost invariably leads to rebound viremia in infected individuals due to a long-lived latent reservoir of integrated proviruses. Therefore, ART must be administered on a life-long basis.
