AMYPAD-PNHS: A Pan-European, Multi-Site & Multimodal MRI Dataset of Older Adults Without Dementia
Abstract The Amyloid Imaging to Prevent Alzheimer’s Disease Prognostic and Natural History Study (AMYPAD-PNHS) multimodal magnetic resonance imaging (MRI) dataset provides open-access longitudinal MRI data of 2759 cognitively normal or mild cognitive impairment individuals, encompassing (micro-)structural, physiological, and functional MRI sequences from 10 European parent cohorts.
Attitudes of specialist memory-clinic patients with early symptomatic Alzheimer’s disease towards lecanemab: results from a multicenter survey in Europe
Abstract Lecanemab approval in the European Union (EU) was granted after a delay. This delay resulted in concerns from many stakeholders, but attitudes of patients with early symptomatic Alzheimer’s disease receiving specialist memory-clinic care remained insufficiently assessed. Therefore, we evaluated attitudes of specialist memory-clinic patients with early symptomatic Alzheimer’s disease towards lecanemab in Europe.
Joint trajectories of brain atrophy, white matter hyperintensities and cognition quantify brain maintenance
|
Abstract Brain maintenance – the preservation of brain structure or function relevant to cognitive performance – remains challenging to quantify.

Smartphone-based detection of subtle memory decline in prodromal Alzheimer’s disease
|
Abstract The slow progression of Alzheimer’s disease (AD) poses a challenge for the quantification of early disease-driven cognitive decline. Here, we show that frequently administered remote and unsupervised digital cognitive assessments can detect differences in cognitive decline within 30 weeks in early AD.

A Clinician View of the EMA Explanation of the Approval of Amyloid-Targeting Therapies
The publication on the approval process of lecanemab and donanemab for early symptomatic Alzheimer’s disease (AD) by members of the Committee for Medicinal Products for Human Use (CHMP) of the European Medicines Agency (EMA) is of great value given the controversies around it.1 Their support for AD research is highly commendable.
Microglia protein profiles in CSF across Alzheimer’s disease clinical stages
|
Abstract Microglia are implicated in the progression of Alzheimer’s disease (AD) pathology from its earliest stages, suggesting that cerebrospinal fluid (CSF) microglia profiling across clinical AD stages can aid in treatment development and monitoring. We analyzed two CSF cohorts (n = 834) that span from unimpaired controls to preclinical and dementia AD stages, identifying 109 dysregulated microglia-related proteins.

Attention in subjective cognitive decline - Authors' reply
I thank Linpei Jia and Hongliang Zhang for their interest in our Personal View on management approaches for the growing group of individuals who seek medical help because of purely subjective cognitive decline (SCD). As research progresses, knowledge about the warning signs of declining cognition in people with SCD will increase.

Refining the understanding of typical Alzheimer disease
Understanding the heterogeneity of typical Alzheimer disease (AD) is of increasing importance for the development of new and effective treatments. A new study in patients with AD described two robust cognitive subtypes with different pathology distributions, genetic backgrounds and clinical trajectories. References 1. Egan, M. F. et al.
