MALT1 protease inhibition restrains glioblastoma progression by reversing tumor-associated macrophage-dependent immunosuppression in mice
Abstract MALT1 protease is an intracellular signaling molecule that promotes tumor progression via cancer cell-intrinsic and cancer cell-extrinsic mechanisms. MALT1 has been mostly studied in lymphocytes, and little is known about its role in tumor-associated macrophages. We show that MALT1 is expressed in glioblastoma (GBM)-associated macrophages.

Translation of Intraoperative Molecular Imaging for Clinical Use
ACTIVE TARGETING The next generation of FGS agents—active tracers—target specific epitopes on tissues of interest. Pafolacianine is a near-infrared molecule that binds to folate receptors α and β, which are highly expressed by a variety of tumors.

NK cells link immune-checkpoint blockade immunotherapy and response in melanoma brain metastases
Natural killer - NK Immune Checkpoint Inhibitor Central Nervous System Cancer Brain metastases (BMs) are the most common intracranial malignancies in adults, affecting 20%–40% of patients with advanced cancer, and markedly outnumbering the incidence of primary brain tumors. Of these, melanoma is one of the tumor types with a high propensity to spread to the brain.

We endeavored to assess both inter- and intra-tumor TAM heterogeneity, by assembling a cohort that spanned glioma grades and molecular subtypes. We profiled TAMs from patient biopsies, derived from 13 untreated primary gliomas (11 glioblastomas [GBMs], two LGGs; Additional file 1: Table S1), and combined this with public data from an additional 580 glioma cases. We performed scRNA-seq on seven of the in-house cases (five GBMs, two LGGs) [10, 11].
