Abstract Penile carcinoma (PeCa) is a rare malignancy occurring in men. Treatment typically involves penectomy, radiation, or cisplatin-based chemotherapy. A paclitaxel, ifosfamide, and cisplatin-based regimen achieves response rates around 65%. However, non-responders face a 5-year survival rate of only 8%, and due to its rarity, it is mediocrely characterized. Thus, an in-depth delineation of PeCa is crucial to identify novel therapeutic targets.

Abstract Somatic mutations in hematopoietic stem/progenitor cells (HSPCs) can lead to clonal hematopoiesis of indeterminate potential (CHIP) and progression to myelodysplastic syndromes (MDS). Using single-cell and anatomical profiling of a large cohort of human bone marrow (BM), we show that the HSPC BM niche in CHIP and MDS is undergoing inflammatory remodeling.

Abstract Cardiac fibroblasts (CF) are key players after myocardial infarction (MI), but their signaling is only incompletely understood. Here we report a first secretome atlas of CF in control (cCF) and post-MI mouse hearts (miCF), combining a rapid cell isolation technique with SILAC and click chemistry. In CF, numerous paracrine factors involved in immune homeostasis are identified. Comparing secretome, transcriptome (SLAMseq), and cellular proteome disclose protein turnover.
