Transcriptional regulator SATB1 limits CD8+ T cell population expansion and effector differentiation in chronic infection and cancer - Nature Immunology
Abstract CD8+ T cells are major mediators of antiviral and antitumor immunity. During persistent antigen stimulation as in chronic infection and cancer, however, they differentiate into exhausted T cells that display impaired functionality. Precursors of exhausted T (TPEX) cells exhibit stem-like properties, including high proliferative, self-renewal and developmental potential, and are responsible for long-term CD8+ T cell responses against persistent antigens.
Lymph nodes fuel KLF2-dependent effector CD8+ T cell differentiation during chronic infection and checkpoint blockade - Nature Immunology
Abstract Exhausted CD8+ T (TEX) cell responses are maintained by precursors of exhausted T (TPEX) cells that possess high self-renewal and developmental potential. TPEX cells also drive the proliferative burst of effector T cells upon therapeutic immune checkpoint blockade (ICB). However, the spatial context and signals that regulate their differentiation and function are not well defined.
β1 integrin signaling governs necroptosis via the chromatin-remodeling factor CHD4
Highlights • Myofibroblasts are the major cell type expressing RIPK3 in induced liver fibrosis • Loss of profibrotic β1 integrin leads to epigenetic repression of Ripk3 by CHD4 • CHD4 represses the Ripk3 locus in a NuRD- and ChAHP-independent manner • The stand-alone function of CHD4 exhibits exquisite locus specificity Summary Fibrosis, characterized by sustained activation of myofibroblasts and excessive extracellular matrix (ECM) deposition, is known to be associated with chronic...
