Cattle and human organoids reveal 2.3.4.4b H5N1 cross-species transmission potential and neuraminidase-specific neutralizing antibodies in humans
|
Abstract The unexpected circulation of clade 2.3.4.4b H5N1 influenza viruses in dairy cattle and the transmission to diverse mammalian species poses a pandemic risk. We sought to explore cattle and human respiratory susceptibility to the 2.3.4.4b H5N1 virus. We establish long-term expandable cattle airway and mammary organoids. The 2.3.4.4b H5N1 virus exhibits high replicative fitness in cattle mammary organoids, recapitulating its remarkable mammary tropism.

Human angiotensin‑converting enzyme 2‑specific benzothiazole-based allosteric inhibitor against pan‑sarbecoviruses
|
Abstract Emerging SARS‑CoV‑2 variants and related zoonotic sarbecoviruses rely on ACE2 for cell entry, motivating host‑directed antivirals that block spike-ACE2 interaction. Here, we characterize MB‑32, a benzothiazole small molecule that binds ACE2, selectively disrupts binding of SARS‑CoV‑2 spike receptor‑binding domain to ACE2, and preserves ACE2 enzymatic activity across species.

YL1004 is a SARS-CoV-2 papain-like protease inhibitor with immunomodulatory and antiviral activity in mice - Nature Communications
Abstract The evolution of SARS-CoV-2, coupled with its immune evasion mechanisms, underscores the urgent need for antiviral strategies beyond vaccination. The papain-like protease (PLpro) is a dual-functional enzyme essential for viral replication and suppression of host innate immunity. Here, we present the rational design and characterization of YL1004, a tricyclic oral PLpro inhibitor demonstrating robust, cross-variant SARS-CoV-2 antiviral activity and favorable pharmacokinetic properties.
Potent and broadly neutralizing antibodies against sarbecoviruses elicited by single ancestral SARS-CoV-2 infection - Communications Biology
Abstract The emergence of various SARS-CoV-2 variants presents challenges for antibody therapeutics, emphasizing the need for more potent and broadly neutralizing antibodies. Here, we employed an unbiased screening approach and successfully isolated two antibodies from individuals with only exposure to ancestral SARS-CoV-2.
COVID-19 drug discovery and treatment options - Nature Reviews Microbiology
Abstract The coronavirus disease 2019 (COVID-19) pandemic caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has caused substantial morbidity and mortality, and serious social and economic disruptions worldwide. Unvaccinated or incompletely vaccinated older individuals with underlying diseases are especially prone to severe disease. In patients with non-fatal disease, long COVID affecting multiple body systems may persist for months.
PMI-controlled mannose metabolism and glycosylation determines tissue tolerance and virus fitness - Nature Communications
Abstract Host survival depends on the elimination of virus and mitigation of tissue damage. Herein, we report the modulation of D-mannose flux rewires the virus-triggered immunometabolic response cascade and reduces tissue damage. Safe and inexpensive D-mannose can compete with glucose for the same transporter and hexokinase.
Potent and broadly neutralizing antibodies against sarbecoviruses induced by sequential COVID-19 vaccination - Cell Discovery
Abstract The current SARS-CoV-2 variants strikingly evade all authorized monoclonal antibodies and threaten the efficacy of serum-neutralizing activity elicited by vaccination or prior infection, urging the need to develop antivirals against SARS-CoV-2 and related sarbecoviruses. Here, we identified both potent and broadly neutralizing antibodies from a five-dose vaccinated donor who exhibited cross-reactive serum-neutralizing activity against diverse coronaviruses.

High-throughput screening of genetic and cellular drivers of syncytium formation induced by the spike protein of SARS-CoV-2 - Nature Biomedical Engineering
Abstract Mapping mutations and discovering cellular determinants that cause the spike protein of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) to induce infected cells to form syncytia would facilitate the development of strategies for blocking the formation of such cell–cell fusion.
Engineered Intranasal Virus Trap Provides Effective Protection Against SARS‐CoV‐2 Infection in Hamsters
1 Introduction While shifting to the endemic stage of Coronavirus Disease 2019 (COVID-19), governments have loosened the regulations on infectious disease control. As societies return to normality, masks are no longer worn in public areas in many countries. As attitudes of the general population have changed, less tests would be done and fewer cases would be reported.
Engineered Intranasal Virus Trap Provides Effective Protection Against SARS‐CoV‐2 Infection in Hamsters
Intranasal Application of Fn-LCB1 Reduces SARS-CoV2 Delta Transmission To test the prophylactic effectiveness of Fn-LCB1 in vivo, we performed a SARS-CoV-2 Delta noncontact transmission experiment with the Golden Syrian hamster model. The Delta variant was used as it showed 100% transmission without protection in our transmission condition. For the experimental setting, we subdivided each hamster cage with an acrylic board.
Vaccine-induced protection against SARS-CoV-2 requires IFN-γ-driven cellular immune response - Nature Communications
Abstract The overall success of worldwide mass vaccination in limiting the negative effect of the COVID-19 pandemics is inevitable, however, recent SARS-CoV-2 variants of concern, especially Omicron and its sub-lineages, efficiently evade humoral immunity mounted upon vaccination or previous infection. Thus, it is an important question whether these variants, or vaccines against them, induce anti-viral cellular immunity.
A new generation Mpro inhibitor with potent activity against SARS-CoV-2 Omicron variants - Signal Transduction and Targeted Therapy
Abstract Emerging SARS-CoV-2 variants, particularly the Omicron variant and its sublineages, continually threaten the global public health. Small molecule antivirals are an effective treatment strategy to fight against the virus. However, the first-generation antivirals either show limited clinical efficacy and/or have some defects in pharmacokinetic (PK) properties. Moreover, with increased use of these drugs across the globe, they face great pressure of drug resistance.
Analogous comparison unravels heightened antiviral defense and boosted viral infection upon immunosuppression in bat organoids - Signal Transduction and Targeted Therapy
Abstract Horseshoe bats host numerous SARS-related coronaviruses without overt disease signs. Bat intestinal organoids, a unique model of bat intestinal epithelium, allow direct comparison with human intestinal organoids. We sought to unravel the cellular mechanism(s) underlying bat tolerance of coronaviruses by comparing the innate immunity in bat and human organoids. We optimized the culture medium, which enabled a consecutive passage of bat intestinal organoids for over one year.
Coronaviruses exploit a host cysteine-aspartic protease for replication - Nature
Abstract Highly pathogenic coronaviruses including severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2)1,2, Middle East respiratory syndrome coronavirus (MERS-CoV)3, and SARS-CoV-14 vary in their transmissibility and pathogenicity. However, infection by all three viruses results in substantial apoptosis in cell culture5-7 and in patient tissues8-10, suggesting a potential link between apoptosis and pathogenesis of coronaviruses.
A broadly neutralizing antibody protects Syrian hamsters against SARS-CoV-2 Omicron challenge - Nature Communications
Abstract The strikingly high transmissibility and antibody evasion of SARS-CoV-2 Omicron variants have posed great challenges to the efficacy of current vaccines and antibody immunotherapy. Here, we screen 34 BNT162b2-vaccinees and isolate a public broadly neutralizing antibody ZCB11 derived from the IGHV1-58 family.

Comparative tropism, replication kinetics, and cell damage profiling of SARS-CoV-2 and SARS-CoV with implications for clinical manifestations, transmissibility, and laboratory studies of COVID-19: an observational study
Summary Background Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) was reported from China in January, 2020. SARS-CoV-2 is efficiently transmitted from person to person and, in 2 months, has caused more than 82 000 laboratory-confirmed cases of coronavirus disease 2019 (COVID-19) and 2800 deaths in 46 countries. The total number of cases and deaths has surpassed that of the 2003 severe acute respiratory syndrome coronavirus (SARS-CoV).
Middle East respiratory syndrome coronavirus and bat coronavirus HKU9 both can utilize GRP78 for attachment onto host cells
Coronavirus tropism is predominantly determined by the interaction between coronavirus spikes and the host receptors. In this regard, coronaviruses have evolved a complicated receptor-recognition system through their spike proteins. Spikes from highly related coronaviruses can recognize distinct receptors, whereas spikes of distant coronaviruses can employ the same cell-surface molecule for entry.
Dual-functional peptide with defective interfering genes effectively protects mice against avian and seasonal influenza
Influenza defective interfering PB2 (DI-PB2), DI-PB1, and DI-PA genes with large internal deletion were generated from the backbone of A/WSN/1933(H1N1) virus using fusion PCR. Each DI-PB2, DI-PB1, and DI-PA consisting of internal deletions were inserted into the phw2000 plasmid (Fig. 1a). When these plasmids of DI-PB2, DI-PB1, and DI-PA were co-transfected or individually transfected into 293T and A549 cells, 7–8 log copies per well of each DIG RNA was detected by RT-qPCR (Fig.
