Lipoyl deglutarylation by ABHD11 regulates mitochondrial and T cell metabolism - Nature Chemical Biology
Abstract Glutarate is an intermediate of amino acid catabolism and an important metabolite for reprogramming T cell immunity. Glutarate exerts its effects either by directly inhibiting metabolite-dependent enzymes or through conjugation to substrates. Intriguingly, glutarylation can occur on protein and nonprotein substrates, but our understanding of these distinct glutaryl modifications is in its infancy.

ABHD11 mediated deglutarylation regulates the TCA cycle and T cell metabolism
Abstract Glutarate is an intermediate of amino acid catabolism and an important metabolite for reprogramming T cell immunity, exerting its effects by inhibition of histone demethylase enzymes or through glutarylation. However, how distinct glutarate modifications are regulated is unclear. Here, we uncover a deglutarylation pathway that couples amino acid catabolism to tricarboxylic acid (TCA) cycle function.

In this issue, Singh et al. show that cofactor LGP2 nucleates MDA5 filaments at internal sites on dsRNA in an ATP-dependent manner. Shown here is an AlphaFold model of LGP2 (yellow) nucleating an MDA5 filament (green).
The constitutively active form of a key cholesterol synthesis enzyme is lipid droplet-localized and upregulated in endometrial cancer tissues
Abstract Cholesterol is essential for both normal cell viability and cancer cell proliferation. Aberrant activity of squalene monooxygenase (SM, also known as squalene epoxidase or SQLE), the rate-limiting enzyme of the committed cholesterol synthesis pathway, is accordingly implicated in a growing list of cancers. We previously reported that hypoxia triggers the truncation of SM to a constitutively active form, thus preserving sterol synthesis during oxygen shortfalls.
Asters: rising stars in the cholesterol universe - Nature Metabolism
Intracellular trafficking of cholesterol is essential for its uptake, storage and export. In this issue of Nature Metabolism, Xiao et al. provide powerful evidence for the importance of hepatic GRAMD1/Aster transporters in maintaining systemic cholesterol homeostasis. Where would we be without cholesterol? Our single most abundant lipid species is vital, yet lethal in excess. Accordingly, we have evolved an elaborate myriad of mechanisms to keep it in check.

Post-translational control of the long and winding road to cholesterol
Laura J. Sharpe, Hudson W. Coates and Andrew J. Brown* School of Biotechnology and Biomolecular Sciences, University of New South Wales, Sydney, New South Wales, Australia ↵*For correspondence: Andrew J. Brown, aj.brown{at}unsw.edu.au. Edited by Dennis R. Voelker Abstract The synthesis of cholesterol requires more than 20 enzymes, many of which are intricately regulated. Post-translational control of these enzymes provides a rapid means for modifying flux through the pathway.
