Previously, we reported that TAGLN2 is highly expressed in immune-related tissues, such as thymus, spleen, and lymph nodes21. Moreover, it is constitutively expressed in primary T (CD3+, CD4+, and CD8+) and B (CD19+) cells (Fig. 1a). In contrast to the lymphoid cells, we noticed that macrophages derived from either bone marrow or peritoneum express minimum levels of TAGLN2 (Fig. 1a and Supplementary Fig. S1a and b), suggesting that it has a minimal function in resting macrophages.

Indoleamine 2,3-dioxygenase (IDO) mediates immune tolerance, and suppressor of cytokine signaling 3 (SOCS3) negatively regulates the JAK/STAT signal transduction pathway. We determined previously that platelet-activating factor (PAF) protects mice against LPS-induced endotoxic shock, but its detailed mechanism of action was unknown. We performed survival experiments in IDO+/+ and IDO?/? mice using an LPS-induced endotoxemia model and rated organ injury (neutrophil infiltration and liver function).

Author contributions: KTN designed, performed and analzed the experiments shown in Fig 1,2, 3 and 6, and and wrote the paper. IDJ designed, performed and analzed the experiments shown in Fig 4,5, and 7. GSC provided technical assistance and contributd to the design and preparion of the figures. MKH and YMP conceived and coordinated the study and wrote the paper. All authors reviewed the results and approved the final version of the manuscript.
