Reversing enhancer RNA–mediated IKBKE gene repression enables synthetic anticancer immunity in prostate cancer models
Abstract Immunotherapy has been effective in many cancer types but has failed in multiple clinical trials in prostate cancers, with the underlying mechanisms remaining largely unclear. Here, we demonstrate that androgen receptor pathway inhibitor (ARPI) plus irradiation (IR) triggered robust anticancer immunity in prostate cancers in both patients and mice.

CHD1 status drives divergent metabolic pathways in SPOP-mutant prostate cancer - Nature Cancer
CHD1 loss is common in SPOP-mutant prostate cancer; however, this combined phenotype has not yet been clearly defined. Genetically engineered mouse models of prostate cancer with Chd1 loss and Spop mutation now reveal a mechanism of castration resistance driven by enhanced intracellular cholesterol and androgen biosynthesis.
Plasma exchange and radiation resensitize immunotherapy-refractory melanoma: a phase I trial - Nature Communications
Abstract Immune checkpoint inhibitors (ICI) are effective for advanced melanoma. However, most develop ICI resistance. Tumor-derived soluble PD-L1 (sPD-L1) and other immunosuppressive factors drive resistance. We hypothesized that therapeutic plasma exchange (TPE) may remove sPD-L1 from circulation and overcome ICI resistance. Patients with metastatic ICI-resistant melanoma and elevated sPD-L1 received radiotherapy to a minority of metastatic lesions, TPE, and ICI re-challenge.
A real-world experience of pembrolizumab monotherapy in microsatellite instability-high and/or tumor mutation burden-high metastatic castration-resistant prostate cancer: outcome analysis | Prostate Cancer and Prostatic Diseases
Abstract The efficacy of pembrolizumab monotherapy in metastatic castration-resistant prostate cancer patients (mCRPC) when stratified by MSI-H and/or TMB-H is poorly defined. Additionally, outcomes based on sequencing source (i.e., tissue or liquid biopsy) have not been well described. We sought to assess outcomes of pembrolizumab monotherapy in patients with mCRPC and compare efficacy based on MSI-H and/or TMB-H when identified by tissue or liquid biopsy.
The yin and yang of chromosomal instability in prostate cancer - Nature Reviews Urology
Abstract Metastatic prostate cancer remains an incurable lethal disease. Studies indicate that prostate cancer accumulates genomic changes during disease progression and displays the highest levels of chromosomal instability (CIN) across all types of metastatic tumours. CIN, which refers to ongoing chromosomal DNA gain or loss during mitosis, and derived aneuploidy, are known to be associated with increased tumour heterogeneity, metastasis and therapy resistance in many tumour types.
Hidden tricks in MATH: Hypermorphic mutations in SPOP tumor suppressor explained by cryo-EM
The mechanism underlying endometrial cancer mutations of SPOP are incompletely understood—the mutated residues seem solvent-exposed in available structures. Cuneo, O’Flynn et al. report the cryo-EM structure of SPOP, which reveals that these mutations are in previously uncharacterized interfaces. Mutations enhance assembly or alter quaternary structure, contributing to super-physiological function.

Microenvironment-Mediated Resistance to Anti-Androgen Therapy
In this issue of Cancer Cell, Zhang et al. expose new possibilities for targeting castration-resistant prostate cancer (CRPC). Activation of the HER2/HER3 axis by cancer-associated fibroblast-secreted NRG1 mediates castration resistance, recommending novel applications of clinical anti-NRG1/HER3 therapeutics in treating CRPC.
