Background The gut microbiome has been linked to responses to immune checkpoint inhibitors (ICIs), yet longitudinal multi-omics studies remain limited. Methods Here, we serially collected gut microbiome data and performed multi-omics analyses, including RNA sequencing (RNA-seq) and whole-exome sequencing (WES), on samples from 92 patients with advanced non-small cell lung cancer (NSCLC) treated with pembrolizumab, to identify factors contributing to treatment response.

Abstract EGFR exon 20 insertion mutations and HER2 alterations are oncogenic drivers in non–small cell lung cancer (NSCLC) with limited therapeutic options. YH42946 is an orally available tyrosine kinase inhibitor designed to selectively target these alterations while sparing EGFR wild-type. Here, we evaluated the in vitro and in vivo antitumor efficacy of YH42946.

Phase I trial of CJRB-101 plus pembrolizumab in patients with metastatic non-small cell lung cancer, head and neck squamous cell carcinoma and melanoma

Only a subset of patients responds to immunotherapy in advanced solid tumors, including non-small cell lung cancer. Modulating the gut microbiome may alleviate resistance to immunotherapy. The trial showed that CJRB-101 plus pembrolizumab yielded an objective response in 58% with progression-free survival of 9 months in immune checkpoint inhibitor (ICI)-naïve metastatic non-small cell lung cancer (NSCLC) with programmed death-ligand 1 >50%.
