Rescuing anxiety by targeting the neuronal-astrocytic iron signaling axis in male mice
Abstract Anxiety disorders affect more than one-quarter of the global population, but current first-line therapies suffer from slow onset and limited efficacy, highlighting the urgent need for novel pathogenic targets. Psychological stress disrupts iron homeostasis in the brain and alters neural circuit excitability; however, the mechanism underlying these two processes remains elusive.
Ferroptosis in kidney disease
Abstract Kidney disease is a growing global public health concern that is associated with high rates of severe morbidity and mortality. Accumulating preclinical and clinical evidence suggests that ferroptosis, an iron-dependent form of regulated cell death characterized by lethal lipid peroxidation, drives the pathogenesis and progression of diverse kidney disorders, including acute kidney injury, chronic kidney disease, renal cell carcinoma and autosomal-dominant polycystic kidney disease.
Mitochondrial fission factor senses and governs ferroptosis
Abstract Ferroptosis is a form of regulated cell death driven by iron-dependent lipid peroxidation and is implicated in diverse pathological states1,2,3,4. Although mitochondria and other organelles are increasingly being recognized as important modulators of ferroptosis5,6,7, a unifying mechanism that couples organelle dynamics to ferroptotic execution has remained elusive.
Zinc accumulation in macrophages exacerbates metabolic dysfunction-associated steatotic liver disease progression
Abstract Metabolic dysfunction-associated steatotic liver disease (MASLD) affects over 1 billion people globally, yet the role of trace elements in its progression remains poorly defined. Here, we identify “macrophage zinc trapping” (i.e., selective zinc accumulation in hepatic macrophages despite systemic zinc depletion) as a critical driver of MASLD. In a cohort of 1069 individuals with MASLD, we found that serum zinc levels were inversely correlated with the disease severity.
Ferroptosis in T cells: mechanisms, biology and translational opportunities
Abstract Ferroptosis is an iron-dependent form of regulated cell death driven by disrupted iron homeostasis and uncontrolled lipid peroxidation. Various metabolites and enzymes regulate cellular sensitivity to ferroptosis by affecting iron, lipid and redox metabolism. These pathways not only signal ferroptotic cell death but also affect the biology of T cells.
Manganese: biology, physiology and role in disease
Abstract Manganese (Mn) has lingered in the shadows as a mere enzymatic cofactor, with its profound role in regulating the most fundamental life processes largely overlooked. This review heralds a “manganese renaissance” — a paradigm shift that elevates Mn from a passive trace element to a dynamic architect of metabolic homeostasis and a critical driver of disease. We synthesize breakthroughs that redefine its biological significance.

TET1 as a master regulator controlling GPX4-dependent and -independent ferroptosis surveillance in acute myeloid leukemia - Nature Communications
Abstract Ferroptosis, an iron-dependent, lipid peroxidation-driven programmed cell death, holds substantial promise for cancer therapy, yet its translational potential is hindered by widespread intrinsic resistance. While glutathione peroxidase 4 (GPX4) is a well-established ferroptosis suppressor, the epigenetic circuitry coordinating GPX4-related mechanisms remains elusive.
SLC7A11 is an unconventional H+ transporter in lysosomes
Keywords lysosome pH optimum acidification cystine ferroptosis Introduction To have an acidic lumen optimal for hydrolase activity, lysosomes maintain a ∼500-fold proton concentration gradient across their membranes.1,2 This gradient also enables the formation of secondary gradients of ions and metabolites that regulate lysosomal functions.3,4 These conditions are required for multiple lysosomal functions, including degradation, cargo delivery, and catabolite export.4,5,6,7 Improper lysosomal...

Iron homeostasis and ferroptosis in human diseases: mechanisms and therapeutic prospects - Signal Transduction and Targeted Therapy
Abstract Iron, an essential mineral in the body, is involved in numerous physiological processes, making the maintenance of iron homeostasis crucial for overall health. Both iron overload and deficiency can cause various disorders and human diseases. Ferroptosis, a form of cell death dependent on iron, is characterized by the extensive peroxidation of lipids.
Characterization of ferroptosis-triggered pyroptotic signaling in heart failure - Signal Transduction and Targeted Therapy
Abstract Pressure overload–induced cardiac hypertrophy is a common cause of heart failure (HF), and emerging evidence suggests that excessive oxidized lipids have a detrimental effect on cardiomyocytes. However, the key regulator of lipid toxicity in cardiomyocytes during this pathological process remains unknown.
Programmable melanoma-targeted radio-immunotherapy via fusogenic liposomes functionalized with multivariate-gated aptamer assemblies - Nature Communications
Abstract Radio-immunotherapy exploits the immunostimulatory features of ionizing radiation (IR) to enhance antitumor effects and offers emerging opportunities for treating invasive tumor indications such as melanoma. However, insufficient dose deposition and immunosuppressive microenvironment (TME) of solid tumors limit its efficacy.
Mechanisms and regulation of defensins in host defense - Signal Transduction and Targeted Therapy
Abstract As a family of cationic host defense peptides, defensins are mainly synthesized by Paneth cells, neutrophils, and epithelial cells, contributing to host defense. Their biological functions in innate immunity, as well as their structure and activity relationships, along with their mechanisms of action and therapeutic potential, have been of great interest in recent years.
Metformin potentiates nephrotoxicity by promoting NETosis in response to renal ferroptosis - Cell Discovery
Abstract Given the rapidly aging population, aging-related diseases are becoming an excessive burden on the global healthcare system. Metformin has been shown to be beneficial to many age-related disorders, as well as increase lifespan in preclinical animal models. During the aging process, kidney function progressively declines. Currently, whether and how metformin protects the kidney remains unclear.
