Abstract Type 2 inflammation is coordinated by remarkably durable CD4+ T helper 2 (TH2) cell responses, yet the cellular architecture that drives this chronic inflammation and prevents exhaustion remains poorly understood. To define the TH2 landscape in chronic type 2 inflammation, we established a mouse model of long-term pulmonary allergen exposure, finding that type 2 inflammation was broadly sustained over time and included an expanded T cell factor 1-expressing progenitor-like population.

Abstract Determining spatial location of cells within tissues gives vital insight into the interactions between resident and inflammatory cells and is a critical factor for uncoupling the mechanisms driving disease. Here, we apply single-cell spatial transcriptomics to reveal the airway wall landscape in health and during asthma. We identified proinflammatory cellular ecosystems that exist within discrete spatial niches in healthy and asthma samples.

Abstract Background The three cysteinyl leukotrienes (cysLTs), (LT)C 4, LTD 4, and LTE 4, have different biological half-lives, cellular targets, and receptor specificities. CysLT 2R binds LTC 4 and LTD 4 in vitro with similar affinities, but displays a marked selectivity for LTC 4 in vivo.
