High-resolution chromatin mapping reveals that CTCF anchors meiotic loops to the chromosome axis
Abstract When precursor germ cells enter meiosis, chromosomes condense into an array of chromatin loops. Using Hi-C and Micro-C, we explored the trajectory of chromatin reorganization from mouse spermatogonia to spermatocytes at the highest temporal and spatial resolution to date. We show that meiotic chromatin reorganization, characterized by a shift from mitotic-to-meiotic cohesins, precedes the traditionally defined meiotic entry point (meiotic G1/S-phase).
Corrigendum: The evolutionary turnover of recombination hot spots contributes to speciation in mice
This extract was created in the absence of an abstract. Genes & Development 30: 266–280 (2016) In the above-mentioned article, the CASTf × 13Rm data set that is presented originated from the wrong mouse strain, likely due to a mix-up during …
High resolution maps of chromatin reorganization through mouse meiosis reveal novel features of the 3D meiotic structure
Abstract When germ cells transition from the mitotic cycle into meiotic prophase I (MPI), chromosomes condense into an array of chromatin loops that are required to promote homolog pairing and genetic recombination. To identify the changes in chromosomal conformation, we isolated nuclei on a trajectory from spermatogonia to the end of MPI. At each stage along this trajectory, we built genomic interaction maps with the highest temporal and spatial resolution to date.

Extensive sex differences at the initiation of genetic recombination
a, SSDS coverage is a measure of DMC1-bound ssDNA either side of each meiotic DSB. In a population of meiocytes, DSBs will occur in a several hundred nucleotide window around the hotspot centre (orange rectangle). To assess coverage, we first convert the position of each SSDS fragment into the distance along the ssDNA from the hotspot centre.
Recombination initiation maps of individual human genomes
The dramatic events of meiotic recombination culminate in the exchange of genetic information between parental chromosomes and ensure the production of genetically distinct gametes. Recombination is initiated by the formation of programmed DNA double-strand breaks (DSBs), and most DSBs occur at discrete hotspots defined by the DNA binding specificity of the PRDM9 protein.
