Decoding stereospecific metabolic regulation of protein modifications
Metabolites can regulate protein structure and function via covalent modifications, with different isomers exerting distinct effects. Now, stereospecific O-2-hydroxyglutarylation by oncometabolites D2HG and L2HG has been uncovered on distinct proteomes, revealing a chiral layer of post-translational regulation that tunes kinase signalling.
Molecular insights into DDX3X–androgen receptor mRNA regulation via non-canonical G-quadruplex in castration-resistant prostate cancer
Abstract Prostate cancer (PC) is one of the most common malignancies in men, and the emergence of androgen receptor-low/negative castration-resistant PC (ARL/– CRPC) following androgen receptor signaling inhibitor (ARSI) therapy remains a critical clinical challenge. The RNA-binding protein DEAD-box helicase 3 X-linked (DDX3X) has been implicated in the translational regulation of androgen receptor (AR) mRNA; however, the underlying binding mechanisms are not well defined.
Enzymatic and Chemical Synthesis for ADP-Ribosylation Using NAD + as Building Blocks: New Concerns in Reaction Discovery and Design
1 Introduction As an important redox coenzyme in cells, NAD+ has expanded its functions from the traditional regulation of energy metabolism to core roles in signal transduction and biomacromolecule modification [1]. ADP-ribosylation, as a typical modification reaction involving NAD+, regulates a series of key physiological processes such as DNA repair, gene expression, and apoptosis by transferring ADP-ribosyl groups to target molecules (e.g., proteins, DNA, and RNA) [2, 3].

Rapid discovery of cyclic peptide protein aggregation inhibitors by continuous selection - Nature Chemical Biology
Abstract Protein aggregates are associated with numerous diseases. Here we report a platform for the rapid phenotypic selection of protein aggregation inhibitors from genetically encoded cyclic peptide libraries in Escherichia coli based on phage-assisted continuous evolution (PACE). We developed a new PACE-compatible selection for protein aggregation inhibition and used it to identify cyclic peptides that suppress amyloid-β42 and human islet amyloid polypeptide aggregation.

A Biomimetic Human Disease Model of Bacterial Keratitis Using a Cornea-on-a-Chip System
A Biomimetic Human Disease Model of Bacterial Keratitis Using a Cornea-on-a-Chip System Bacterial keratitis is a common form of inflammation caused by the bacterial invasion of the corneal stroma after trauma. In extreme cases, it can lead to severe visual impairment or even blindness; therefore, timely medical intervention is imperative. Abstract Lysine l-lactylation (Kl-la) is a novel protein posttranslational modification (PTM) driven by l-lactate. This PTM has three isomers: Kl-la, N-ε-(carboxyethyl)-lysine (Kce) and d-lactyl-lysine (Kd-la), which are often confused in the context of the Warburg effect and nuclear presence.
The subcommissural organ regulates brain development via secreted peptides - Nature Neuroscience
Abstract The subcommissural organ (SCO) is a gland located at the entrance of the aqueduct of Sylvius in the brain. It exists in species as distantly related as amphioxus and humans, but its function is largely unknown. Here, to explore its function, we compared transcriptomes of SCO and non-SCO brain regions and found three genes, Sspo, Car3 and Spdef, that are highly expressed in the SCO.

With the advent of new energy vehicles, smart grids, and portable electronic devices in today's society, researchers have begun to focus on safer and cheaper energy storage technologies [1]. The elevated energy density of lithium-ion batteries (LIBs) makes them the most commonly used accumulator devices in electric vehicles and various electronic devices [2], [3], [4]. However, the scarcity, high cost, and uneven distribution of lithium resources limit its wider applications.

Integrating intracellular and extracellular proteomic profiling for in‐depth investigations of cellular communication in a model of prostate cancer
4 DISCUSSION A critical aspect of systems biology research is understanding how cellular communication and signaling networks operate across model types [30]. Peptidomics - Nature Reviews Methods Primers
Abstract Peptides are biopolymers, typically consisting of 2–50 amino acids. They are biologically produced by the cellular ribosomal machinery or by non-ribosomal enzymes and, sometimes, other dedicated ligases. Peptides are arranged as linear chains or cycles, and include post-translational modifications, unusual amino acids and stabilizing motifs. Their structure and molecular size render them a unique chemical space, between small molecules and larger proteins.
