Expression of progranulin (GP88) protein appears as an independent prognostic factor for clinical progression in high-risk prostate cancer patients.
Prostate cancer (PCa), the second most common cancer, is still a major cause of morbidity and mortality among men worldwide.
Expression of progranulin (GP88) protein appears as an independent prognostic factor for clinical progression in high-risk prostate cancer patients
Abstract Prostate cancer (PCa), the second most common cancer, is still a major cause of morbidity and mortality among men worldwide.
Deep phenotyping of skin tissue remodeling in patients with systemic sclerosis treated with CD19-CAR T cells
|
Abstract Systemic sclerosis (SSc) is an autoimmune disease characterized by vasculopathy and fibrotic remodeling of the skin and internal organs. Fibrotic tissue changes are considered hardly reversible with current therapies, suggesting that new strategies are required to modulate the disease-associated molecular and cellular phenotype to enable regeneration of affected tissues.

A multimodal dataset for precision oncology in head and neck cancer - Nature Communications
Abstract Head and neck cancer is a common disease and is associated with a poor prognosis. A promising approach to improving patient outcomes is personalized treatment, which uses information from a variety of modalities. However, only little progress has been made due to the lack of large public datasets. We present a multimodal dataset, HANCOCK, that comprises monocentric, real-world data of 763 head and neck cancer patients.
Effects of different B-cell-depleting strategies on the lymphatic tissue
WHAT IS ALREADY KNOWN ON THIS TOPIC • Advanced protein-based B cell depleters, such as glycoengineered CD20 monoclonal antibody obinutuzumab (OBI) and CD19/CD3 T cell engager blinatumomab (BLI), are promising new therapeutic instruments for treatment of autoimmune disease (AIDs). It has been speculated that such B cell-depleting modalities achieve better clearance of tissue B cells in patients.
BCMA-CAR T-cells in a patient with relapsing idiopathic inflammatory myositis after CD19-CAR T-cells - Nature Medicine
Abstract CD19 chimeric antigen receptor (CD19-CAR) T-cell therapy has been shown to induce stable drug-free remission in patients with refractory autoimmune disease. The management of potential relapses is currently unclear. Here, we report on a 45-year-old woman with treatment-refractory Jo-1-associated anti-synthetase syndrome, who initially achieved disease remission after CD19-CAR T-cell therapy but then experienced disease relapse after 9 months.

Tumor budding and lymphovascular invasion as prognostic factors in p16-positive oropharyngeal squamous cell carcinomas - British Journal of Cancer
Abstract We aimed to validate the prognostic significance of tumor budding (TB) in p16-positive oropharyngeal squamous cell carcinomas (OPSCC). We analyzed digitized H&E-stained slides from a multicenter cohort of five large university centers consisting of n = 275 cases of p16-positive OPSCC.

CD19-CAR T-cell therapy induces deep tissue depletion of B cells
Patients and methods We analysed inguinal lymph node biopsies from five patients undergoing CD19-CAR T-cell therapy. Biopsies were performed before CD19-CAR T-cell therapy (before the conditioning therapy) and after CD19-CAR T-cell therapy (before the reappearance of B cells) from an inguinal lymph node at baseline and from the homolateral region, following topographic and ultrasound landmarks identified during the previous biopsy, at follow-up.
Neutrophil extracellular traps characterize caseating granulomas - Cell Death & Disease
Abstract Tuberculosis (TB) remains one of the top 10 causes of death worldwide and still poses a serious challenge to public health. Recent attention to neutrophils has uncovered unexplored areas demanding further investigation. Therefore, the aim of this study was to determine neutrophil activation and circulatory neutrophil extracellular trap (NET) formation in various types of TB.

Bispecific T cell engager therapy for refractory rheumatoid arthritis - Nature Medicine
Abstract Bispecific T cell engagers (BiTEs) kill B cells by engaging T cells. BiTEs are highly effective in acute lymphoblastic leukemia. Here we treated six patients with multidrug-resistant rheumatoid arthritis (RA) with the CD19xCD3 BiTE blinatumomab under compassionate use. Low doses of blinatumomab led to B cell depletion and concomitant decrease of T cells, documenting their engager function.

Endocytoscopy for assessing histologic inflammation in ulcerative colitis: development and prospective validation of the ELECT (ErLangen Endocytoscopy in ColiTis) score (with videos)
Background and Aims Apart from endoscopic healing as an established treatment goal in patients with inflammatory bowel disease (IBD), histologic remission is an emerging endpoint that might even better predict disease outcome, especially in ulcerative colitis (UC). Within this study, we aimed to evaluate whether endocytoscopy (EC) as an in vivo contact microscopy technology can accurately assess histologic inflammation and predict the further course of disease in UC patients.
pT3 colorectal cancer revisited: a multicentric study on the histological depth of invasion in more than 1000 pT3 carcinomas-proposal for a new pT3a/pT3b subclassification | British Journal of Cancer
Abstract Background Pathological TNM staging (pTNM) is the strongest prognosticator in colorectal carcinoma (CRC) and the foundation of its post-operative clinical management. Tumours that invade pericolic/perirectal adipose tissue generally fall into the pT3 category without further subdivision. Methods The histological depth of invasion into the pericolic/perirectal fat was digitally and conventionally measured in a training cohort of 950 CRCs (Munich).
Prognostic and Predictive Value of Fibroblast Growth Factor Receptor Alterations in High-grade Non-muscle-invasive Bladder Cancer Treated with and Without Bacillus Calmette-Guérin Immunotherapy - European Urology
Abstract Background Limited data are available on the prognostic and predictive value of fibroblast growth factor receptor alterations (FGFRa) relative to clinical outcomes in patients with non–muscle-invasive bladder cancer (NMIBC). Objective To determine whether FGFRa may be clinically useful in stratifying for treatment response in a real-world cohort of patients with pT1 NMIBC treated and untreated with bacillus Calmette-Guérin (BCG) instillation therapy.

Deep Learning Predicts Molecular Subtype of Muscle-invasive Bladder Cancer from Conventional Histopathological Slides
Abstract Background Muscle-invasive bladder cancer (MIBC) is the second most common genitourinary malignancy, and is associated with high morbidity and mortality. Recently, molecular subtypes of MIBC have been identified, which have important clinical implications. Objective In the current study, we tried to predict the molecular subtype of MIBC samples from conventional histomorphology alone using deep learning.

Piwi-like 1 and -2 protein expression levels are prognostic factors for muscle invasive urothelial bladder cancer patients
Piwi-like 1/-2 expression and correlation with clinico-pathological parameters and expression of selected proteins We studied a cohort of 95 MIBC for their Piwi-like 1 and Piwi-like 2 protein expression by immunohistochemistry (IHC). The clinico-pathological data of the MIBC patients are summarized in Table 1. Piwi-like 1/-2 protein expression was detected in the cytoplasm and assessed in an IRS score. Table 1 Clinico-pathological data for MIBC patients.
