RGS22 is a metazoa-specific radial spoke component required for coordinated ciliary beating
Abstract In metazoan epithelia, arrays of motile cilia beat in a tissue-wide coordinated manner to drive unidirectional fluid flow. Whether radial spokes (RSs), which mediate mechano-signal transduction between the central pair and axonemal dynein arms, acquire metazoa-specific subunit(s) in evolution to facilitate this function remains poorly known. Here we report that Gα-GTPase activating protein family member RGS22 is a metazoa-specific component of RS1/2 complexes.
Loss-of-function variants in ODAD1 disrupt ODA docking and induce actin cytoskeletal remodeling in primary ciliary dyskinesia
Abstract Primary ciliary dyskinesia (PCD) is a genetically heterogeneous disorder characterized by defective mucociliary clearance due to impaired motile cilia function. Outer dynein arm docking complex subunit 1 (ODAD1) encodes a structural component of the outer dynein arm docking complex (ODA-DC), which is crucial for effective ciliary beating. However, the full spectrum of ODAD1-associated pathogenesis remains incompletely defined.
Doublet microtubule-associated tektins and enzymes differentially regulate sperm flagellar integrity and motility - Nature Communications
Abstract Doublet microtubule (DMT)-associated proteins assemble and drive sperm flagella, which are essential for successful fertilization. However, the exact roles of different DMT-associated proteins in sperm function and the underlying molecular mechanisms remain elusive. Here, we generate four gene-knockout mice based on high-resolution structures targeting distinct DMT components: two intermediate filament-like tektins (TEKT1, TEKT5) and two enzymes (TSSK6, DUSP21).
Mammalian PIWI–piRNA–target complexes reveal features for broad and efficient target silencing - Nature Structural & Molecular Biology
Abstract The PIWI-interacting RNA (piRNA) pathway is an adaptive defense system wherein piRNAs guide PIWI family Argonaute proteins to recognize and silence ever-evolving selfish genetic elements and ensure genome integrity. Driven by this intensive host–pathogen arms race, the piRNA pathway and its targeted transposons have coevolved rapidly in a species-specific manner, but how the piRNA pathway adapts specifically to target silencing in mammals remains elusive.
Axonemal structures reveal mechanoregulatory and disease mechanisms - Nature
Abstract Motile cilia and flagella beat rhythmically on the surface of cells to power the flow of fluid and to enable spermatozoa and unicellular eukaryotes to swim. In humans, defective ciliary motility can lead to male infertility and a congenital disorder called primary ciliary dyskinesia (PCD), in which impaired clearance of mucus by the cilia causes chronic respiratory infections1.
Two-Dimensional Modelling of Sludge Heat and Mass Transfer in a Paddle Dryer
Abstract : In this paper, we propose a concise but general two-dimensional model based on the penetration model in order to simulate the heat and mass transfer and drying kinetics of sludge in the paddle dryer. The sludge control equations were developed to simulate the heat and mass transfer of sludge and the penetration model was introduced to describe the mixing of sludge.

Ciliary central apparatus structure reveals mechanisms of microtubule patterning - Nature Structural & Molecular Biology
Abstract A pair of extensively modified microtubules form the central apparatus (CA) of the axoneme of most motile cilia, where they regulate ciliary motility. The external surfaces of both CA microtubules are patterned asymmetrically with large protein complexes that repeat every 16 or 32 nm. The composition of these projections and the mechanisms that establish asymmetry and longitudinal periodicity are unknown.
De novo identification of mammalian ciliary motility proteins using cryo-EM
Highlights • Structure of the 48-nm repeat of a mammalian doublet microtubule from respiratory cilia • Tektin filaments bind within the lumens of doublet microtubules • A pentameric docking complex attaches axonemal dyneins to doublet microtubules • Loss of connectivity across microtubule walls causes ciliopathy-like phenotypes Summary Dynein-decorated doublet microtubules (DMTs) are critical components of the oscillatory molecular machine of cilia, the axoneme, and have luminal surfaces...
Cryo-EM structure of an activated GPCR-G protein complex in lipid nanodiscs
Abstract G-protein-coupled receptors (GPCRs) are the largest superfamily of transmembrane proteins and the targets of over 30% of currently marketed pharmaceuticals. Although several structures have been solved for GPCR–G protein complexes, few are in a lipid membrane environment. Here, we report cryo-EM structures of complexes of neurotensin, neurotensin receptor 1 and Gαi1β1γ1 in two conformational states, resolved to resolutions of 4.1 and 4.2 Å.
Structures of radial spokes and associated complexes important for ciliary motility
Abstract In motile cilia, a mechanoregulatory network is responsible for converting the action of thousands of dynein motors bound to doublet microtubules into a single propulsive waveform. Here, we use two complementary cryo-EM strategies to determine structures of the major mechanoregulators that bind ciliary doublet microtubules in Chlamydomonas reinhardtii. We determine structures of isolated radial spoke RS1 and the microtubule-bound RS1, RS2 and the nexin−dynein regulatory complex (N-DRC).
