Valproate independently activates Snf1, inhibits TORC1, and induces repression of INO1 transcription by increasing nuclear localization of Opi1 - Scientific Reports
Abstract Valproate (VPA), a drug used to treat neurological disorders such as epilepsy, bipolar disorder, and migraines, induces inositol depletion in the brain by preventing its synthesis, which is a hypothesized therapeutic mechanism for mood stabilizing drugs. However, the mechanism by which this occurs is not known. VPA treatment reduces activity of the enzyme that catalyzes the rate-limiting step of inositol synthesis, myo-inositol-3-phosphate synthase (MIPS).

Phosphatidic acid inhibits inositol synthesis by inducing nuclear translocation of kinase IP6K1 and repression of myo-inositol-3-P synthase
Introduction Inositol is a six-carbon cyclitol that is essential for all eukaryotic life ( 1 Case K.C. Salsaa M. Yu W. Greenberg M.L. Regulation of Inositol Biosynthesis: Balancing Health and Pathophysiology. ). Deprivation of inositol results in “inositol-less death”, a phenotype originally described in yeast and supported by early work in human cell lines ( 2 Keith A.D. Pollard E.C. Snipes W. Inositol-less death in yeast results in a simultaneous increase in intracellular viscosity.

One can only imagine how a list like this could be used.

The Vitality of Students for Public Sociology
“Post-truth” populism and rising inequality have led to a resurgence of public scholarship. Universities are struggling to reassert their public mission and fight off accusations of elitism and irrelevance. Scholars, appalled by the lurch towards nativism and anti-intellectualism, have re-committed to partnering with local communities to tackle increasingly entrenched social problems. Meanwhile, communities are in greater need of support from researchers and public institutions than ever before.

No Place Like Home: Housing crisis breeds opportunity
Crises, the saying goes, create opportunities. By forcing deep problems into the light of day, crises can focus our attention on their origins and impacts, and inspire us to work collectively to build transformative solutions. The affordable-housing crisis in Santa Cruz and across California presents us with just this kind of opportunity. Our UCSC-based No Place Like Home project arose three years ago to heed this call.

Inositol Depletion Induced by Acute Treatment of the Bipolar Disorder Drug Valproate Increases Levels of Phytosphingosine
Bipolar disorder (BD) is a severe psychiatric illness affecting ?1% of the world population. Valproate (VPA) and lithium, widely used for the treatment of BD, are not universally effective. These drugs have been shown to cause inositol depletion, but translating this observation to a specific therapeutic mechanism has been difficult, hampering the development of more effective therapies.

Inositol Hexakisphosphate Kinase 1 (IP6K1) Regulates Inositol Synthesis in Mammalian Cells
myo-Inositol, the precursor of all inositol compounds, has pivotal roles in cell metabolism and signaling pathways. Although physiological studies indicate a strong correlation between abnormal intracellular inositol levels and neurological disorders, very little is known about the regulation of inositol synthesis in mammalian cells.
Inositol depletion, GSK3 inhibition and bipolar disorder
Valproic acid and lithium are widely used to treat bipolar disorder, a severe illness characterized by cycles of mania and depression. However, their efficacy is limited, and treatment is often accompanied by serious side effects. The therapeutic mechanisms of these drugs are not understood, hampering the development of more effective treatments. Among the plethora of biochemical effects of the drugs, those that are common to both may be more related to therapeutic efficacy.
