Results MSNs were derived in vitro from iPSCs using a previously established protocol, which yields highly enriched MSN cultures (23; Figure 1A). The iPSC lines were obtained from two unrelated healthy individuals (Con-1, Con-2), three unrelated individuals carrying the C9ORF72RE mutation (C9-1, C9-2, C9-3), and one gene-edited line (C9-Δ3), created from C9-3 using CRISPR-Cas9 technology to remove the GGGGCC repeat expansion mutation.

Although Greatwall represents a potential oncogene and prospective therapeutic target, our understanding of the cellular and molecular consequences of chemical Greatwall inactivation remains limited.

Abstract The rifamycin synthetase (RIFS) from the bacterium Amycolatopsis mediterranei is a large (3.5 MDa) multienzyme system that catalyzes over 40 chemical reactions to generate a complex precursor to the antibiotic rifamycin B. It is considered a hybrid enzymatic assembly line and consists of an N-terminal nonribosomal peptide synthetase loading module followed by a decamodular polyketide synthase (PKS).
