Olfactory Receptor Activation Reduces Platelet Reactivity and Arterial Thrombosis Through Actin Cytoskeleton Remodeling
Research Article Open Access Clinical Perspective • A platelet OR (olfactory receptor) was identified as a novel, druggable target for reducing high residual platelet reactivity. • Platelet OR2L13 activation by a nonodorant chemical probe CCF0054500 broadly inhibits platelet reactivity through PAR1, P2Y12, GPVI (glycoprotein VI), and thromboxane receptors, as well as GPIIb/IIIa, uniquely inhibiting both biochemical and biomechanical activation of the platelet.

Oral Immunotherapy Should Play a Key Role in Preschool Food Allergy Management
Favourable preschool OIT safety and effectiveness data supports its key role in food allergy management. Awaiting natural resolution may confer a higher risk of severe reactions than previously thought. Future research should prioritise confirming the potential positive impact of preschool OIT on QoL. 1 Introduction Food allergy is a common, potentially life-threatening condition that usually begins in early childhood and often does not resolve throughout an individual's lifetime.
Protein Sequence Modelling with Bayesian Flow Networks
Abstract Exploring the vast and largely uncharted territory of amino acid sequences is crucial for understanding complex protein functions and the engineering of novel therapeutic proteins. Whilst generative machine learning has advanced protein sequence modelling, no existing approach is proficient for both unconditional and conditional generation. In this work, we propose that Bayesian Flow Networks (BFNs), a recently introduced framework for generative modelling, can address these challenges.

Naples adopts a former table grape as its darling white wine variety
NAPLES -- When you think of Naples, you probably don’t think of wine. You may think of pizza…or even fried pizza. If you are ill-informed, you may think of nothing more than a corrupt, crime-ridden city, calling for secretly sewn pockets to conceal valuables throughout your piumini and pantaloni. What one should think, simply, purely, is life.
Franciacorta: value artisanal sparkling hidden by the shadow of Champagne
ROME - You have been duped by Champagne. Let’s face it: our society is driven by consumerism and the perceived value behind what we are consuming. A presumption of prestige or poverty is found behind every label. Champagne, in this sense, is luxury. Champagne is a romance. Champagne is myth. In fact, the myth of its creation floated through circles of the cognoscenti for decades until very recently.
My man on the Janiculum: In pursuit of Michael Mewshaw via Graham Greene
ROME -- Who is Michael Mewshaw? Even in the ever-shrinking world of literati, a man as storied and prolific in publication as Mewshaw is far from adequately known, much less championed. It’s not surprising that his idol and mentor, Graham Greene, was in some ways, equally absent from the cognescenti. Late-era “Boomers” fawn over Greene – arguably the greatest British author of the Twentieth Century with a name that should float off the tongue in the same breath as Hemingway, or Fitzgerald.
Is Amazon polluting the Chesapeake Bay?
Is the company named for South America's greatest river and rainforest hurting America's greatest estuary, the Chesapeake Bay? Amazon and its fellow data center operators Google, Meta and Microsoft - all based in the environmentally conscious Seattle and San Francisco areas - are driving a huge and growing demand for solar energy in Virginia to power their expanding network of Northern Virginia data centers.

Azithromycin possesses biofilm-inhibitory activity and potentiates non-bactericidal colistin methanesulfonate (CMS) and polymyxin B against Klebsiella pneumonia | PLOS ONE
Loading metrics Open Access Peer-reviewed Research Article Abstract Novel antibiotic combinations may act synergistically to inhibit the growth of multidrug-resistant bacterial pathogens but predicting which combination will be successful is difficult, and standard antimicrobial susceptibility testing may not identify important physiological differences between planktonic free-swimming and biofilm-protected surface-attached sessile cells.
Throwing Money at Infrastructure Projects Is Not Enough
When Congress passed the bipartisan Infrastructure Investment and Jobs Act in late 2021, the floor debate referenced “shovel-ready” projects that would promptly produce results. Unfortunately, the hope of shovel-ready projects isn’t realistic. Even modest budget increases can be hard for the bureaucracy to digest, and when the increases are massive, delays can seem interminable. We need to invest in the workforce needed to effectively implement the infrastructure law.

by John Salustri The industrial market nationally “continues to be the darling of commercial real estate,” reports JLL in its Q2 2021 Outlook. “On the leasing front, things are moving so quickly that buildings are not sitting idle or vacant for long. Availability in the market is extremely tight, with all time low vacancies and new market-high rents.” Compared to the current woes of the office or retail sectors, that’s good news for landlords and investors.

New Insights Into the Mechanisms of Thrombosis Formation: A Focus on Inflammation
Venous thromboembolism (VTE) represents a leading cause of global mortality. Most recent clinical studies have focused on improving patient mortality using advanced hemodynamic parameters as risk stratification tools and then advanced interventions beyond the use of systemic anticoagulation, with mixed results.1,2 It is now clear that the long-term consequences of residual pulmonary thrombosis and thrombus burden in patients with acute pulmonary embolism (PE) should be evaluated.
Stimuli-responsive Pd2L4 metallosupramolecular cages: towards targeted cisplatin drug delivery
Stimuli-responsive Pd2L4 metallosupramolecular cages: towards targeted cisplatin drug delivery† Metallosupramolecular cages are an emerging, but as of yet relativity unexplored, drug delivery vector. Herein we show that discrete dipalladium(II) molecular cages of the formula [Pd2L4](X)4 can be quantatively self-assembled from a simple tripyridyl ligand (2,6-bis(pyridin-3-ylethynyl)pyridine) and [Pd(CH3CN)4](X)2 (X = BF4− or SbF6−).

Loyalty Lost? Your Best Customers Are Cheating On You
By Scott Cameron Many retailers are in the dark about how loyal their best customers are. Yes, they realize that those customers do buy at other stores, but they don’t know how much they spend there. New findings reveal that many retailers’ best customers (defined here as the top 20 percent of customers by spend) aren’t really all that loyal. In fact, they spend a lot at competitors. This “lost-sales” issue stems from a flawed assumption: that retailers’ “best customers” shop mostly with them.

Thousands of students, teachers, parents and victims carried signs and marched across Illinois in the cold, snow and sleet on Saturday as part of the nationwide March for Our Lives against gun violence. The marches were largely led by students, many of them moved to action after last month’s high school shooting in Parkland, Florida. At least 23 marches were scheduled across the state, from Woodstock to Carbondale.
Intracellular rebinding of transition-state analogues provides extended in vivo inhibition lifetimes on human purine nucleoside phosphorylase
Purine nucleoside phosphorylase (PNP) is part of the human purine salvage pathway. Its deficiency triggers apoptosis of activated T-cells, making it a target for T-cell proliferative disorders. Transition-state analogues of PNP bind with picomolar (pm) dissociation constants. Tight-binding PNP inhibitors show exceptionally long lifetimes on the target enzyme. We solve the mechanism of the target residence time by comparing functional off-rates in vitro and in vivo.
