Abstract Pattern-recognition receptor (PRR) agonists are valuable agents across multiple medical applications, from vaccinology to immune-oncology. However, well-defined and potent small molecule agonists for many PRRs still await discovery and development. Screening of chemical libraries of ~200,000 small molecules for maturation of human monocytic cells by quantifying NF-κB activation and cell adherence was completed.

Abstract Human whole blood (WB) immunophenotyping may represent the in vivo immunological state with better fidelity than artificially isolated peripheral blood mononuclear cells (PBMCs). We used a deep phenomics modeling approach to elucidate the quantitative differences in major immune cell lineages in WB and PBMC compartments in a steady-state in vitro setting. We studied functional innate immune responses induced by pattern recognition receptor agonist adjuvants (PRRa).

1 Purpose and Appropriate Sample Types This 17-parameter panel was designed and optimized for the deep immunophenotyping and functional characterization of T cell subsets in murine spleen (Table 2 and Table ). Functional cytokines (IFNγ, TNF, and IL-2/4/5/17) in the T cell compartment initiate the innate immune responses after infection or immunization and establish a bridge between adaptive immune responses, which ultimately confer long-lasting protective immunity.
