Longitudinal changes in DNA methylation in IDH-mutant glioma fuel disease progression through altered cell state differentiation
Abstract The progression of isocitrate dehydrogenase-mutant glioma (IDH-G) from slow-growing tumor to fatal disease is associated with transcriptional and DNA methylation changes that remain poorly understood. Here, we profiled a longitudinal cohort of 36 IDH-G samples from 19 patients by joint-capture multi-omic single-nucleus DNA methylation, single-nucleus RNA sequencing and bulk exome sequencing.

Machine learning identifies prognosticators of intracranial metastatic disease in patients with breast or lung cancer
Abstract Intracranial metastatic disease is a severe complication of cancer that confers substantial morbidity and mortality. Patients with breast or lung cancer are at particularly elevated risk of IMD. Early identification of individuals at increased risk could enable targeted surveillance and timely intervention. We developed interpretable machine-learning competing-risk models to estimate the risk of intracranial metastatic disease among patients with breast or lung cancer.
AI Augmented Confocal Laser Endomicroscopy for Rapid Intraoperative Diagnosis of Brain Tumors
Abstract Intraoperative diagnosis is important for brain tumor surgery. Although frozen section (FS) remains the standard, its time demands and logistical limitations hinder real-time decision-making. Confocal laser endomicroscopy (CLE) offers rapid high-resolution imaging for efficient diagnosis. This multicenter prospective trial evaluated the diagnostic performance of CLE while developing a novel Swin Transformer–based AI diagnostic model. A total of 461 biopsies from 376 patients were analyzed.
Planar Hall Effect in Quasi-Two-Dimensional Materials
The planar Hall effect in 3D systems is an effective probe for their Berry curvature, topology, and electronic properties. However, the Berry curvature-induced conventional planar Hall effect is forbidden in 2D systems as the out-of-plane Berry curvature cannot couple to the band velocity of the electrons moving in the 2D plane.
Profiling Breast Tumor Heterogeneity and Identifying Breast Cancer Subtypes Through Tumor‐Associated Immune Cell Signatures and Immuno Nano Sensors
1 Introduction Breast cancer is a significant health concern affecting women worldwide, accounting for 12.5% of all newly diagnosed cancers annually.[1] Breast cancer is highly heterogeneous with various cancer cells within the same tumor existing in diverse metabolic states due to variations in cellular, genetic, epigenetic, and molecular expression.[2-4] These molecular expressions are not uniformly constant and can dynamically evolve over time as the tumor progresses to a different...

Chiral anomalies in three-dimensional spin-orbit coupled metals: Electrical, thermal, and gravitational anomalies
Abstract The discovery of chiral anomaly in Weyl semimetals, the nonconservation of chiral charge and energy across two opposite chirality Weyl nodes, has sparked immense interest in understanding its impact on various physical phenomena. Here, we demonstrate the existence of electrical, thermal, and gravitational quantum chiral anomalies in three-dimensional (3D) spin-orbit coupled systems.

Assessing the Association of Targeted Therapy and Intracranial Metastatic Disease
Key Points Question What is the association of targeted therapy with survival for patients with intracranial metastatic disease (IMD)? Findings In this cohort study of 26 676 patients with IMD, prolonged survival was observed in patients with IMD and ERBB2 (formerly HER2)–positive breast cancer, EGFR-positive lung and bronchus cancer, or BRAF-positive melanoma who received targeted therapy compared with those who did not.
Amid COVID, first do no harm
I am a surgeon, mother, daughter, researcher, and ethicist. These multiple roles dancing together to a more discordant tune amid this unprecedented time. The “daughter” has managed to convince her ageing parents that they really ought to not leave the house and that she will step in to accommodate their needs. Simultaneously, the “surgeon” is surprised when her 80-year-old mother, Layla, relays that she received a phone call telling her she needs to present to the hospital for some tests.
Gun control is a Canadian value I was proud to adopt

I was completing my residency training in neurosurgery at Northwestern University and had been recruited to the University of Toronto to start a clinical practice and research program focused on treatment of brain cancers.

ID1 inhibition may improve efficacy of glioblastoma treatment
Investigators from St. Michael’s Hospital and The Hospital for Sick Children in Toronto have determined that the transcriptional regulatory protein ID1 appears to maintain cancer stem cells in glioblastoma. Results from studies conducted in mice — which laid the groundwork for a trial in humans — suggest ID1 inhibition could lead to better treatment efficacy.
