Cancer-independent somatic mutation of the wild-type NF1 allele in normal tissues in neurofibromatosis type 1 - Nature Genetics
Abstract Cancer predisposition syndromes mediated by recessive cancer genes generate tumors via somatic variants (second hits) in the unaffected allele. Second hits may or may not be sufficient for neoplastic transformation. Here we performed whole-genome and whole-exome sequencing on 479 tissue biopsies from a child with neurofibromatosis type 1, a multisystem cancer-predisposing syndrome mediated by constitutive monoallelic NF1 inactivation.

Risk factors for detection of SARS-CoV-2 in healthcare workers during April 2020 in a UK hospital testing programme
Abstract Background: Healthcare workers (HCW) are a crucial part of the workforce but are also at potentially at increased risk of infection from SARS-CoV-2. Emerging evidence has suggested specific groups of HCW are at further increased risk particularly those from Black, Asian and Minority Ethnic (BAME) groups. Previous reports have not examined risk factors associated with contracting the virus and were reported prior to the pandemic peak in the UK.

Cerebral arteriopathy associated with heterozygous variants in the casitas B-lineage lymphoma gene
Abstract Objective To report a series of patients with cerebral arteriopathy associated with heterozygous variants in the casitas B-lineage lymphoma (CBL) gene and examine the functional role of the identified mutant Cbl protein. We hypothesized that mutated Cbl fails to act as a negative regulator of the RAS-mitogen-activated protein kinases (MAPK) signaling pathway, resulting in enhanced vascular fibroblast proliferation and migration and enhanced angiogenesis and collateral vessel formation.

Is chronic lymphocytic inflammation with pontine perivascular enhancement responsive to steroids (CLIPPERS) in children the same condition as in adults? - Sa - - Developmental Medicine & Child Neurology - Wiley Online Library
This case series describes three children with chronic lymphocytic inflammation with pontine perivascular enhancement responsive to steroids (CLIPPERS), an inflammatory condition characterized by a relapsing–remitting disease course responsive to steroids. The patients (two males, age 3y and 13y; one female, age 14y) presented with ataxia, dysarthria, and multiple cranial neuropathies.
Taxonomy of CNS Tumours; A Series of Three Short Reviews on the WHO 2016 Classification and Beyond
In May 2016, an update of the 4th edition of the World Health Organization (WHO) Classification of Tumours of the Central Nervous System (CNS) was officially released [1]. This revised 4th edition represents a paradigm shift as, for the first time, the definition of multiple CNS tumour entities is now partly based on particular genotypic characteristics.
Anti-HMGCR Autoantibodies in Juvenile Idiopathic Inflammatory Myopathies Identify a Rare but Clinically Important Subset of Patients
AbstractObjective. We aimed to establish the prevalence and clinical associations of anti-HMG-CoA-reductase (anti-HMGCR) in a large UK cohort with juvenile myositis.Methods. There were 381 patients investigated for anti-HMGCR using ELISA.Results. Anti-HMGCR autoantibodies were detected in 4 patients (1%). These children had no or minimal rash and significant muscle disease.
MEK inhibition appears to improve symptom control in primary NRAS-driven CNS melanoma in children
Trametinib is the first therapy to show any objective or measurable effect in NRAS -mutated primary CNS melanoma, with few side effects in this small series. The role of this therapy should be explored further in this rare paediatric tumour. All four had an improvement in symptoms and objectively in signs. These varied from mild improvement for 1 month, to a sustained symptom-free period of 9 months in one case.
