A non-invasive end-to-end intelligent assistance system for breast ultrasound
|
Abstract Although artificial intelligence enhances medical image classification to effectively improve lesion diagnosis accuracy and efficiency, it still faces generalization challenges in breast ultrasound and its clinical potential remains underutilized in complex real-world scenarios. Here, we develop and test an end-to-end breast intelligent recognition device (BIRD) for women across diverse institution/population datasets.

Inhibiting hedgehog signal by a patched-1 antibody - Cell Discovery
Dear Editor, The Hedgehog (HH) signaling pathway is crucial for the developmental patterning of animal embryogenesis and the regeneration of adult tissues1. The aberrant activation of HH signaling has been linked to various cancers, such as basal cell carcinoma and medulloblastoma1. Consequently, investigations into HH signaling not only contribute to understanding the molecular mechanisms underlying animal development but also offer valuable insights into potential cancer treatment approaches.
Hydrothermal Synthesis of Stable Lead-Free Cs4MnBi2Cl12 Perovskite Single Crystals for Efficient Photocatalytic Degradation of Organic Pollutants
Hydrothermal Synthesis of Stable Lead-Free Cs4MnBi2Cl12 Perovskite Single Crystals for Efficient Photocatalytic Degradation of Organic Pollutants Xiaofeng Qi, Fei Zhang, Zhipeng Chen, Xu Chen, Mochen Jia, Huifang Ji and Zhifeng Shi Abstract Recently, the photocatalytic degradation of organic pollutants using halide perovskite materials have increasingly attracted people's attention.
Regulated degradation of HMG CoA reductase requires conformational changes in sterol-sensing domain - Nature Communications
Abstract 3-Hydroxy-3-methylglutaryl coenzyme A reductase (HMGCR) is the rate-limiting enzyme in cholesterol synthesis and target of cholesterol-lowering statin drugs. Accumulation of sterols in endoplasmic reticulum (ER) membranes accelerates degradation of HMGCR, slowing the synthesis of cholesterol. Degradation of HMGCR is inhibited by its binding to UBIAD1 (UbiA prenyltransferase domain-containing protein-1).
Mechanisms and inhibition of Porcupine-mediated Wnt acylation - Nature
Abstract Wnt signalling is essential for regulation of embryonic development and adult tissue homeostasis1,2,3, and aberrant Wnt signalling is frequently associated with cancers4. Wnt signalling requires palmitoleoylation on a hairpin 2 motif by the endoplasmic reticulum-resident membrane-bound O-acyltransferase Porcupine5,6,7 (PORCN). This modification is indispensable for Wnt binding to its receptor Frizzled, which triggers signalling8,9.
Cryo-EM structure of oxysterol-bound human Smoothened coupled to a heterotrimeric Gi
Abstract The oncoprotein Smoothened (SMO), a G-protein-coupled receptor (GPCR) of the Frizzled-class (class-F), transduces the Hedgehog signal from the tumour suppressor Patched-1 (PTCH1) to the glioma-associated-oncogene (GLI) transcription factors, which activates the Hedgehog signalling pathway1,2. It has remained unknown how PTCH1 modulates SMO, how SMO is stimulated to form a complex with heterotrimeric G proteins and whether G-protein coupling contributes to the activation of GLI proteins3.
Two Patched molecules engage distinct sites on Hedgehog yielding a signaling-competent complex
The Hedgehog (HH) signaling pathway is important in development, and excessive HH signaling is associated with cancer. Signaling occurs through the G protein–coupled receptor Smoothened. The pathway is repressed by the membrane receptor Patched-1 (PTCH1), and this inhibition is relieved when PTCH1 binds the secreted protein HH. Two recent papers have described structures of HH bound to PTCH1, but surprisingly, each described a different binding epitope on HH. Qi et al.

Structures of human Patched and its complex with native palmitoylated sonic hedgehog
Hedgehog (HH) signalling governs embryogenesis and adult tissue homeostasis in mammals and other multicellular organisms1,2,3. Whereas deficient HH signalling leads to birth defects, unrestrained HH signalling is implicated in human cancers2,4,5,6. N-terminally palmitoylated HH releases the repression of Patched to the oncoprotein smoothened (SMO); however, the mechanism by which HH recognizes Patched is unclear.
